TNF- α promotes CXCL-1/8 production in keratinocytes by downregulating galectin-3 through NF-κB and hsa-miR-27a-3p pathway to contribute psoriasis development
CONCLUSION: TNF-α initiates psoriatic inflammation by increasing CXCL-1/8 in keratinocytes mediated by the axis of NF-κB-hsa-miR-27a-3p-galectin-3 pathway.PMID:37358143 | DOI:10.1080/08923973.2023.2229510

Protein Kinase CK2 Promotes Proliferation, Abnormal Differentiation, and Proinflammatory Cytokine Production of Keratinocytes via Regulation of STAT3 and Akt Pathways in Psoriasis
Am J Pathol. 2023 May;193(5):567-578. doi: 10.1016/j.ajpath.2023.01.016.ABSTRACTProtein kinase CK2 is a constitutively active and ubiquitously expressed serine/threonine kinase that is closely associated with various types of cancers, autoimmune disorders, and inflammation. However, the role of CK2 in psoriasis remains unknown. Herein, the study indicated elevated expression of CK2 in skin lesions from patients with psoriasis and from psoriasis-like mice. In the psoriasis-like mouse model, the CK2-specific inhibitor CX-4945 ameliorated imiquimod-induced psoriasis symptoms with reduced proliferation, abnormal differentiatio...

Suprabasin-derived polypeptides: SBSN(50-63) induces inflammatory response via TLR4-mediated mast cell activation
This study was conducted to clarify whether SBSN(50-63) plays a pivotal role in activating mast cells and mediating proinflammatory cytokines and chemokines production in psoriasis. The increased expression of SBSN in psoriatic lesions was confirmed by bioinformatics analysis, PCR and ELISA. Wild-type mice injected subcutaneously with SBSN(50-63) exhibited infiltration of inflammatory cells and the release of cytokines in vivo. SBSN(50-63) stimulated mouse primary mast cells (MPMC) and the laboratory of allergic disease 2 (LAD2) human mast cells to produce more inflammatory mediators than the control group, which were meas...

Inhibition of serum and glucocorticoid-regulated protein kinase-1 aggravates imiquimod-induced psoriatic dermatitis and enhances proinflammatory cytokine expression via the NF-kappaB pathway
In this study, we found that SGK1 expression was decreased in macrophages from psoriasis patients. Moreover, a specific pharmacological SGK1 inhibitor, EMD638683, significantly enhanced imiquimod (IMQ)-mediated TLR7/8 activity and proinflammatory cytokine production in RAW264.7 cells, and this result was confirmed by SGK1 siRNA.

Neuroblast Differentiation-Associated Protein Derived Polypeptides: AHNAK(5758-5775) Induces Inflammation by Activating Mast Cells via ST2
This study aimed to investigate whether AHNAK (5758-5775), a neuroblast differentiation-associated protein-derived polypeptide, could be considered a new endogenous substance in psoriasis patients, which activates mast cells and induces the skin inflammatory response contributing to psoriasis. Wild-type mice were treated with AHNAK(5758-5775) to observe the infiltration of inflammatory cells in the skin and cytokine release in vivo. The release of inflammatory mediators by mouse primary mast cells and the laboratory of allergic disease 2 (LAD2) human mast cells was measured in vitro. Molecular docking analysis, molecular d...

374 Eukaryotic translation initiation factor 4E (eIF4E) as a target of anti-psoriatic treatment
Eukaryotic translation initiation factor 4E (eIF4E) has been known to play a critical role in the regulation of gene expression in eukaryotes and to affect many essential cellular processes, including proliferation, apoptosis and differentiation. We and others found eIF4E and other eIF subunits upregulated in human psoriatic skin and thus hypothesized that they are involved in the pathophysiology of psoriasis and can serve as direct topical targets for anti-psoriatic treatment. We used specific eIF4E siRNA or briciclib an eIF4E inhibitor in HaCaT cells, a human 3D-psoriasis tissue model as well as the imiquimod and TGF ß ...
