Activated Mast Cells Mediate Low-Grade Inflammation in Type 2 Diabetes: Interleukin-37 Could be Beneficial
Publication date: Available online 31 January 2018Source: Canadian Journal of DiabetesAuthor(s): Pio Conti, Gianpaolo Ronconi, Spyridon K. Kritas, Alessandro Caraffa, Theoharis C. TheoharidesAbstractMast cells (MCs) promote guest immune responses to parasites and play a critical role in allergic and inflammatory reactions. Once they have been activated, MCs release highly inflammatory compounds that can provoke serious pathologic signs that can lead to death. MCs generate a number of preformed, de novo synthesized compounds and inflammatory cytokine/chemokine synthesis in response to the high-affinity (Kd=10–10 M) immuno...

Long-term risk of rosiglitazone on cardiovascular events: a systematic review and meta-analysis.
Abstract Rosiglitazone has been proposed as a treatment strategy for type 2 diabetes mellitus (T2DM), could provide robust glucose-lowering capability with risk of cardiovascular events. We thus did a systematic review and meta-analysis of controlled trials to assess the effect of this treatment on glycaemic control, cardiovascular events in patients with T2DM. We systematically search PubMed, Embase and Cochrane Central Register of Controlled Trials comparing rosiglitazone to other anti-diabetic treatments. These studies included randomized controlled trials (RCTs), cohort studies, and case-control studies that h...

Leptin, cardiovascular diseases and type 2 diabetes mellitus.
Abstract Leptin, an adipokine that is implicated in the control of food intake via appetite suppression, may also stimulate oxidative stress, inflammation, thrombosis, arterial stiffness, angiogenesis and atherogenesis. These leptin-induced effects may predispose to the development of cardiovascular diseases. In the present review we discuss the evidence linking leptin levels with the presence, severity and/or prognosis of both coronary artery disease and non-cardiac vascular diseases such as stroke, carotid artery disease, peripheral artery disease (PAD) and abdominal aortic aneurysms (AAA) as well as with chroni...

Combination of SGLT-2 inhibitors and GLP-1 receptor agonists: potential benefits in surrogate and hard endpoints.
CONCLUSION: The combination of metformin with SGLT2i, GLP-1 RA, and a potent statin, in high CVD risk patients with DM, is expected to substantially reduce CVD mortality and morbidity, improving the quality of life of patients with DM at the same time. Prospective studies are needed to confirm this finding. PMID: 29865997 [PubMed - as supplied by publisher]

Preparation And Evaluation Of Eudragit Coated Chitosan Microparticles For Oral Insulin Delivery
Objectives Diabetes mellitus was known in relic and vestiges at present a global and ascalating health problem with millions of cases being diagnosed daily. Diabetes can affect numerous parts of the body and is associated with solemn complications, such as heart disease and stroke, blindness, kidney failure, and lower-limb amputation. Some complications, particularly micro vascular (e.g., eye, kidney, and nerve) disease, can be reduced with good glucose control.

Causal Impact of Type 2 Diabetes Mellitus on Cerebral Small Vessel Disease Clinical Sciences
Conclusions—Our Mendelian randomization study provides evidence to suggest that T2D may be causally associated with CSVD, in particular with lacunar stroke and FA.

O5.7. risk of diabetic complications and subsequent mortality among individuals with schizophrenia and diabetes mellitus: a nationwide population-based register study
DiscussionUnexpectedly, we found individuals with comorbid schizophrenia and diabetes mellitus to have a similar or lower rate of diabetic complications diagnosed in hospitals compared to individuals with diabetes mellitus only. However, we still found an excess mortality following a diagnosis of a diabetic complication among individuals with schizophrenia. These results may indicate that individuals are not even seen in hospitals with their diabetic complications and hence indicate an increased need for improved somatic care of individuals with schizophrenia if the burden of diabetes mellitus morbidity and mortality should be reduced.

Circulating Branched-Chain Amino Acids and Incident Cardiovascular Disease in a Prospective Cohort of US Women Original Articles
Conclusions: Circulating plasma BCAAs were positively associated with incident CVD in women. Impaired BCAA metabolism may capture the long-term risk of the common cause underlying T2D and CVD.

Correlation between acute stroke-induced white matter lesions and insulin resistance
The present study was to examine the relationship between white matter lesions (WMLs) and insulin resistance (IR) in patients with acute stroke and evaluate clinical prognosis. Around 200 patients with initial onset of acute stroke including 146 patients (73.0%) with WMLs and 54 patients (27%) without WMLs were analyzed by neuropsychological scales. Fasting blood glucose (FBG), fasting insulin, blood lipid, homocysteine (Hcy), high-sensitivity C-reactive protein (hs-CRP), creatinine, and uric acid, diabetes mellitus (DM), prediabetes (PD), and normal glucose (NG) were determined according to HbA1c levels. According to hom...

Lifestyle interventions for improving health and health behaviours in women with type 2 diabetes: A systematic review of the literature 2011 –2017
Type 2 diabetes mellitus (T2DM) results from the body ’s ineffective use of insulin [1] and while it is often associated with ‘Western lifestyle’ [2], the prevalence of T2DM is rising fastest in low- and middle-income countries [3]. Global estimates suggest that more than 422 million people are living with diabetes [3], with T2DM accounting for 9 0% of cases [4]. Diabetes also increases the risk of other comorbid conditions. For example, a recent systematic review of more than 100 prospective studies showed that people with diabetes had a 2–3 fold increased risk of vascular diseases like coronary heart disease and ischemic stroke [5].

Sodium-glucose cotransporter 2 inhibitors and ischemic stroke.
CONCLUSION: Despite the multiple pleiotropic effects of SGLT-2 inhibitors, these agents do not appear to affect stroke risk. Ongoing large trials with longer follow-up will evaluate whether the pleiotropic effects of this class will translate into benefits in ischemic stroke prevention. Letter to the Editor: Pneumocephalus: Is the needle size significant?
Discussion. Pneumocephalus is defined by two mechanisms: a ball-valve and an inverted bottle concept.1 The ball-valve type implies positive pressure events, such as coughing or valsalva maneuvers, that prevent air escape. Tension pneumocephalus is included in this mechanism, causing a parenchymal mass effect. The inverted bottle theory includes a negative intracranial pressure gradient following cerebrospinal fluid drainage, relieved by air influx. A small pneumocephalus is usually sealed by blood clots or granulation, allowing spontaneous reabsorption and resolution.[1] Otherwise, the lateral positioning of a patient duri...

Plasma proprotein ‐convertase‐subtilisin/kexin type 9 (PCSK9) and cardiovascular events in type 2 diabetes
ConclusionsWe found that PCSK9 was inconsistently associated with CV events in populations with type 2 diabetes. The association may depend on the level of CV risk and the background treatment.
