Effects of SOST Gene Silencing on Proliferation, Apoptosis, Invasion, and Migration of Human Osteosarcoma Cells Through the Wnt/ β-Catenin Signaling Pathway
AbstractOur study explored the effects ofSOST gene silencing on the proliferation, apoptosis, invasion, and migration of human osteosarcoma cells through Wnt/ β-catenin signaling pathway. Fresh tissues were obtained from 108 patients with osteosarcoma and 46 patients with osteochondroma. Human osteosarcoma cells (MG-63, U2-OS, HOS, and Saos-2) and normal osteoblast (hFoB1.19) were selected and cultured. Osteosarcoma cells were grouped randomly into the b lank group, the scrambled control group, and theSOST-siRNA group. Cell proliferation was determined by MTT assay. Cell cycle and apoptosis were tested by flow cytometry. ...

Hypoxia ‐induced proliferation of HeLa cells depends on epidermal growth factor receptor‐mediated arginase II induction
Abstract Solid tumors can often be hypoxic in regions, and cancer cells can respond to hypoxia with an increase in proliferation and a decrease in apoptosis, leading to a net increase in viable cell numbers. We have recently found that in an osteosarcoma cell line, hypoxia‐induced proliferation depends on arginase II induction. Epidermal growth factor receptor (EGFR) has been shown to mediate the hypoxia‐induced cellular proliferation in some cancer cell lines. We hypothesized that hypoxia‐induced proliferation of HeLa cells would depend on arginase II induction and that this induction of arginase II would occur thro...

Downregulation of 14-3-3 β inhibits proliferation and migration in osteosarcoma cells.
Downregulation of 14-3-3β inhibits proliferation and migration in osteosarcoma cells. Mol Med Rep. 2017 Nov 22;: Authors: Wu Q, Zhu J, Liu F, Liu J, Li M Abstract The 14-3-3 protein isoform β (14‑3‑3β), which is an intracellular adaptor protein that exists in all eukaryotic organisms, is highly expressed in many cancer tissues, including glioma, lung carcinoma and breast cancer. However, 14‑3‑3β expression and function in osteosarcoma progression remain unknown. In the present study, the endogenous expression of 14‑3‑3β was assessed in osteosarcoma samples and the effect of 14‑3‑3...

LIM kinase 1 serves an important role in the multidrug resistance of osteosarcoma cells.
Authors: Yang JZ, Huang LH, Chen R, Meng LJ, Gao YY, Ji QY, Wang Y Abstract Multidrug resistance (MDR) is a major challenge for the management of the majority of cancers. The precise molecular mechanisms of MDR remain elusive. In a previous study, a multidrug resistant osteosarcoma model [MG63/vincristine (VCR)] was established by intermittent exposure of MG63 cells to gradually increasing concentrations of VCR. These cells exhibited cross-resistance to multiple structurally and mechanistically unrelated chemotherapeutic agents. The development of MDR was associated with increased expression of LIM kinase 1 (LIMK1)...

Anticancer effect of (S)-crizotinib on osteosarcoma cells by targeting MTH1 and activating reactive oxygen species
This study aimed to explore the anticancer effect of MTH1-targeted drug (S)-crizotinib on osteosarcoma (OS) cells. We detected MTH1 expression in OS tissues and cells using immunohistochemistry and western blot. The effects of MTH1 on OS cell viability were explored using the siRNA technique and CCK8. The anticancer effects of the MTH1-targeted drug (S)-crizotinib on OS cells were explored by in-vitro assays. The intracellular 8-oxo-dGTP level and oxygen reactive species (ROS) of OS cells were detected by Cy3-conjugated avidin staining and dichlorofluorescein diacetate staining, respectively. The expression of MTH1 was sig...

GSE120512 Transcriptome modulation by loss-of-YAP1/TAZ in SK-N-MC Ewing Sarcoma cells
Contributors : Juan Diaz-Martin ; Laura Romero-PerezSeries Type : Expression profiling by arrayOrganism : Homo sapiensYAP1 (Yes-associated protein 1) and TAZ (transcriptional coactivator with PDZ-binding motif, or WWTR1) are nucleo-cytoplasmic shuttling proteins that can function in the nucleus as transcriptional coactivators. Here we sought to evaluate which genes are regulated by endogenous levels of YAP/TAZ. We compared SK-N-MC cells transfected with a control non-targeting siRNA with cells transfected with a mix of siRNA targeting both YAP and TAZ.

Trichostatin A activates FOXO1 and induces autophagy in osteosarcoma.
Conclusions: We found that inhibition of autophagy, either by autophagy inhibitors or ATG gene knockdown, markedly enhances TSA-caused cell death. Taken together, our studies reveal the function of autophagy in HDACI-caused osteosarcoma cell death and thus support the development of a novel therapeutic strategy by combining HDACIs and autophagy inhibitors in osteosarcoma treatment. Long noncoding RNA ERICD interacts with ARID3A via E2F1 and regulates migration and proliferation of osteosarcoma cells.
Abstract Long noncoding RNA (lncRNA) dysregulation is known to be taking part in majority of cancers, including osteosarcoma. In one of our previous studies, we showed that lncRNA MEG3 is being regulated by microRNA-664a (miR-664a) suppresses the migratory potential of osteosarcoma cells (U-2OS). We now report a novel lncRNA, namely, ERICD, which is linked to the transcription factor AT-rich interaction domain 3A (ARID3A) in U-2OS cells. We show that ARID3A binds to ERICD and indirectly interacts with each other via the E2F transcription factor 1 (E2F1). Furthermore, small interfering RNA (siRNA)-mediated knockdow...
