Volatile free liquid membranes for electromembrane extraction
Anal Chim Acta. 2021 Oct 16;1182:338959. doi: 10.1016/j.aca.2021.338959. Epub 2021 Aug 16.ABSTRACTVolatile solvents are excellent extraction media for liquid-liquid extractions. However, their use in supported liquid membranes (SLMs) is limited by their evaporation from SLM and thus poor SLM stability and they have never been considered truly useful for electromembrane extraction (EME). In this contribution, volatile solvents were systematically investigated as liquid membranes for EME and their extraction characteristics were comprehensively examined for the first time. A short plug of a water immiscible volatile solvent ...

The effect of smoking on the plasma concentration of tricyclic antidepressants - a systematic review
CONCLUSION: The effect of smoking on the plasma concentration of TCAs is still not entirely clear. There is a possibility that smoking affects the distribution of TCA metabolites, but this is probably not of clinical importance.PMID:34497000 | DOI:10.1017/neu.2021.28 (Source: Acta Neuropsychiatrica)

A Scoping Review of Interventions for Spousal Bereavement in Older Adults
Am J Geriatr Psychiatry. 2021 Aug 16:S1064-7481(21)00423-1. doi: 10.1016/j.jagp.2021.08.002. Online ahead of print.ABSTRACTThe loss of a spouse is a common and natural life event for older adults. Nearly one of four older bereaved spouses experience prolonged grief, impaired function or chronic depression. Mechanisms underlying these and other long-term health risks are not well understood. We conducted a scoping literature review to examine the interventions and outcomes that have been studied for late-life spousal bereavement to date. We identified 22 studies of group and individual-level interventions with most studies ...

Dose-dependent opposite effects of nortriptyline on affective-like behavior in adolescent rats: Comparison with adult rats
In conclusion, clear behavioral and neurochemical differences were observed between adolescent and adult rats in response to nortriptyline treatment. Interestingly, while nortriptyline displayed an antidepressant-like potential at the lowest dose examined in adolescence, a higher dose shifted these results towards a negative outcome, thus reinforcing the need to extreme caution when considering this treatment for our younger population.PMID:34464602 | DOI:10.1016/j.ejphar.2021.174465 (Source: European Journal of Pharmacology)

Tricyclic antidepressants versus ‘active placebo’, placebo or no intervention for adults with major depressive disorder: a protocol for a systematic review with meta-analysis and Trial Sequential Analysis
DiscussionTricyclic antidepressants are recommended by clinical guidelines and frequently used worldwide in the treatment of major depressive disorder. There is a need for a thorough systematic review to provide the necessary background for weighing the benefits against the harms. This review will ultimately inform best practice in the treatment of major depressive disorder.Systematic review registrationPROSPEROCRD42021226161. (Source: Systematic Reviews)

Treatment of Long-term Sudden Sensorineural Hearing Loss as an Otologic Migraine Phenomenon
Objectives: To describe a cohort of patients presenting with long-term sudden sensorineural hearing loss (SSNHL) treated with prophylactic migraine and intratympanic steroid therapy. Methods: Patients presenting to a neurotology clinic at least 6 weeks from SSNHL onset were included. All patients received migraine prophylactic medication (nortriptyline, topiramate, and/or verapamil) and lifestyle changes for at least 6 weeks, as well as intratympanic steroid injections, if appropriate. Results: Twenty-one patients (43% female) with a mean age of 64 ± 11 years who presented 9 ± 8 months (...

Beneficial and harmful effects of antidepressants versus placebo, ‘active placebo’, or no intervention for adults with major depressive disorder: a protocol for a systematic review of published and unpublished data with meta-analyses and trial sequential analyses
DiscussionAs antidepressants are commonly used to treat major depressive disorder in adults, a systematic review evaluating their beneficial and harmful effects is urgently needed. This review will inform best practice in treatment and clinical research of this highly prevalent and burdensome disorder.Systematic review registrationPROSPEROCRD42020220279 (Source: Systematic Reviews)

Effects of Genetic Polymorphism in CYP2D6, CYP2C19, and the Organic Cation Transporter OCT1 on Amitriptyline Pharmacokinetics in Healthy Volunteers and Depressive Disorder Patients
The tricyclic antidepressant amitriptyline is frequently prescribed but its use is limited by its narrow therapeutic range and large variation in pharmacokinetics. Apart from interindividual differences in the activity of the metabolising enzymes cytochrome P450 (CYP) 2D6 and 2C19, genetic polymorphism of the hepatic influx transporter organic cation transporter 1 (OCT1) could be contributing to interindividual variation in pharmacokinetics. Here, the impact of OCT1 genetic variation on the pharmacokinetics of amitriptyline and its active metabolite nortriptyline was studied in vitro as well as in healthy volunteers and in...

Determination of drugs in exhumed liver and brain tissue after over 9  years of burial by liquid chromatography–tandem mass spectrometry—Part 2: Benzodiazepines, opioids, and further drugs
This paper may serve as reference for exhumations with a postmortem interval of more than 9.5 years in which an investigation of benzodiazepines, opioids, and further drugs is performed. The benzodiazepines diazepam, its metabolites, 7 ‐aminoflunitrazepam, and midazolam as well as all opioids (like fentanyl, morphine, and tramadol), and 4‐aminoantipyrine, 4‐methylaminoantipyrine, ambroxol, amitriptyline, its metabolite nortriptyline, bupivacaine, etomidate, glibenclamide, nortriptyline, its metabolite amitriptyline, pantopra zole, promethazine, propofol, rocuronium, and ropivacaine were detectable in liver and brain ...
