BNIP3 in hypoxia-induced mitophagy: Novel insights and promising target for non-alcoholic fatty liver disease
This study aimed to investigate the expression and the role of BNIP3 in NAFLD under hypoxia, and explore its involvement in regulating NAFLD mitophagy, fatty acid β-oxidation both in vivo and in vitro. BNIP3-mediated mitophagy level was analyzed using real-time quantitative polymerase chain reaction, Western blotting, immunofluorescence and electron microscopy. The role of BNIP3 in fatty acid β-oxidation was evaluated using lipid droplet staining, triglyceride content determination, and cellular energy metabolism. The results showed that compared with the HFD-2200 m, the body weight, inflammatory liver injury, and lipid ...

Hedgehog signalling in CD4 < sup > + < /sup > T helper cell polarisation
Int J Biochem Cell Biol. 2024 Mar;168:106518. doi: 10.1016/j.biocel.2024.106518. Epub 2024 Jan 11.ABSTRACTCD4+ T cells are critical in orchestrating immune responses against various pathogens and cancer but can also be drivers of autoimmune disease, allergy and pro-tumour responses. Naïve CD4+ T cells polarise into specialised T helper cell subsets with unique effector functions. While the guiding transcription factors and effector molecules of the T helper cell lineages are well understood, the signalling pathways orchestrating the intricate T helper cell polarisation programmes remain poorly understood. Here we review a...

UBC9 stabilizes PFKFB3 to promote aerobic glycolysis and proliferation of glioblastoma cells
Int J Biochem Cell Biol. 2023 Dec;165:106491. doi: 10.1016/j.biocel.2023.106491. Epub 2023 Oct 28.ABSTRACTCancer cells prefer to utilizing aerobic glycolysis to generate energy and anabolic metabolic intermediates for cell growth. However, whether the activities of glycolytic enzymes can be regulated by specific posttranslational modifications, such as SUMOylation, in response to oncogenic signallings, thereby promoting the Warburg effect, remain largely unclear. Here, we demonstrate that phosphofructo-2-kinase/fructose-2,6-bisphosphatase 3 (PFKFB3), a key glycolytic enzyme, interacts with SUMO-conjugating enzyme UBC9 and ...

Molecular and cellular evidence of a direct interaction between the TRAF2 C-terminal domain and ganglioside GM1
Int J Biochem Cell Biol. 2023 Dec 22;167:106508. doi: 10.1016/j.biocel.2023.106508. Online ahead of print.ABSTRACTTNF receptor-associated factor 2 (TRAF2) is involved in different cellular processes including signal transduction and transcription regulation. We here provide evidence of a direct interaction between the TRAF domain of TRAF2 and the monosialotetrahexosylganglioside (GM1). Previously, we showed that the TRAF domain occurs mainly in a trimeric form in solution, but it can also exist as a stable monomer when in the nanomolar concentration range. Here, we report that the quaternary structure of the TRAF domain is...

The Role of Zinc Finger Proteins in the Fate Determination of Mesenchymal Stem Cells during Osteogenic and Adipogenic Differentiation
Int J Biochem Cell Biol. 2023 Dec 22:106507. doi: 10.1016/j.biocel.2023.106507. Online ahead of print.ABSTRACTZinc finger proteins (ZFPs) constitute a crucial group of transcription factors widely present in various organisms. They act as transcription factors, nucleases, and RNA-binding proteins, playing significant roles in cell differentiation, growth, and development. With extensive research on ZFPs, their roles in the determination of mesenchymal stem cells (MSCs) fate during osteogenic and adipogenic differentiation processes have become increasingly clear. ZFP521, for instance, is identified as an inhibitor of the W...
