Affibody-based hBCMA x CD16 dual engagers for NK cell-mediated killing of multiple myeloma cells
We describe the development and characterization of the (to date) smallest Natural Killer (NK) cell re-directing human B Cell Maturation Antigen (hBCMA) x CD16 dual engagers for potential treatment of multiple myeloma, based on combinations of small 58 amino acid, non-immunoglobulin, affibody affinity proteins. Affibody molecules to human CD16a were selected from a combinatorial library by phage display resulting in the identification of three unique binders with affinities (KD) for CD16a in the range of 100 nM-3 µM. The affibody exhibiting the highest affinity demonstrated insensitivity towards the CD16a allotype (158F/V...

Biomaterials Text Mining: A Hands-On Comparative Study of Methods on Polydioxanone Biocompatibility
N Biotechnol. 2023 Sep 4:S1871-6784(23)00044-4. doi: 10.1016/j.nbt.2023.09.001. Online ahead of print.ABSTRACTScientific information extraction is fundamental for research and innovation, but is currently mostly a manual, time-consuming process. Text Mining tools (TMTs) enable automated, accurate and quick information extraction from text, but there is little precedent of their use in the biomaterials field. Here, we compare the ability of various TMTs to extract useful information from biomaterials abstracts. Focusing on the biocompatibility of polydioxanone, a biodegradable polymer for which there are relatively few scie...

Machine translation of standardised medical terminology using natural language processing: A scoping review
N Biotechnol. 2023 Aug 29;77:120-129. doi: 10.1016/j.nbt.2023.08.004. Online ahead of print.ABSTRACTStandardised medical terminologies are used to ensure accurate and consistent communication of information and to facilitate data exchange. Currently, many terminologies are only available in English, which hinders international research and automated processing of medical data. Natural language processing (NLP) and Machine Translation (MT) methods can be used to automatically translate these terms. This scoping review examines the research on automated translation of standardised medical terminology. A search was performed ...

High throughput purification of monoclonal recombinant antibodies using a Protein-A coated membrane plate system
N Biotechnol. 2023 Aug 28:S1871-6784(23)00042-0. doi: 10.1016/j.nbt.2023.08.003. Online ahead of print.ABSTRACTThe therapeutic use of monoclonal antibodies (mAbs) ranges from cancer treatment to immune-mediated conditions, covering infectious and cardiovascular disorders, among others. The development of improved methods for therapeutic antibody discovery has accelerated the identification of numerous mAbs: a discovery campaign can be deeply mined, resulting in hundreds, even thousands, of potential antibody leads for a given target of interest. High throughput mAb expression and purification methods are required for the r...

Novel GH109 enzymes for bioconversion of group A red blood cells to the universal donor group O
N Biotechnol. 2023 Aug 27:S1871-6784(23)00041-9. doi: 10.1016/j.nbt.2023.08.002. Online ahead of print.ABSTRACTGlycoside hydrolases (GHs) have been employed for industrial and biotechnological purposes and often play an important role in new applications. The red blood cell (RBC) antigen system depends on the composition of oligosaccharides on the surface of erythrocytes, thus defining the ABO blood type classification. Incorrect blood transfusions may lead to fatal consequences, making the availability of the correct blood group critical. In this regard, it has been demonstrated that some GHs may be helpful in the convers...
