S1PR3 agonism and S1P lyase inhibition rescue mice in the severe state of experimental sepsis
Biomed Pharmacother. 2024 Apr 9;174:116575. doi: 10.1016/j.biopha.2024.116575. Online ahead of print.ABSTRACTSepsis is characterized as life-threatening organ dysfunction caused by a dysregulated host response to an infection. Despite numerous clinical trials that addressed this syndrome, there is still no causative treatment available to dampen its severity. Curtailing the infection at an early stage with anti-infectives is the only effective treatment regime besides intensive care. In search for additional treatment options, we recently discovered the inhibition of the sphingosine 1-phosphate (S1P) lyase and subsequent a...

NGF increases Connexin-43 expression and function in pulmonary arterial smooth muscle cells to induce pulmonary artery hyperreactivity
CONCLUSIONS: Modulation of Cx43 by NGF in pulmonary arterial smooth muscle cells contributes to NGF-induced alterations of pulmonary artery reactivity. Since NGF and its TrkA receptor play a role in vivo in Cx43 increased expression in PH induced by chronic hypoxia, these NGF/Cx43-dependent mechanisms may therefore play a significant role in human PH pathophysiology.PMID:38599061 | DOI:10.1016/j.biopha.2024.116552 (Source: Biomedicine and pharmacotherapy = Biomedecine and pharmacotherapie)

Effect of adalimumab as an anti-inflammatory agent on gene expression of retinal pigment epithelial cells
In conclusion, despite its safety, the 2x concentration of ADA was the only dose that did not ignite the expression of any of the studied inflammatory and fibrogenesis genes. This dosage, which is roughly equal to 2 mg intravitreal dose in a clinical setting, might be referred to as a reference starting point for future in-vivo studies in ocular conditions.PMID:38599062 | DOI:10.1016/j.biopha.2024.116568 (Source: Biomedicine and pharmacotherapy = Biomedecine and pharmacotherapie)

Roles and mechanisms of copper homeostasis and cuproptosis in osteoarticular diseases
Biomed Pharmacother. 2024 Apr 9;174:116570. doi: 10.1016/j.biopha.2024.116570. Online ahead of print.ABSTRACTCopper is an essential trace element in the human body that is extensively distributed throughout various tissues. The appropriate level of copper is crucial to maintaining the life activities of the human body, and the excess and deficiency of copper can lead to various diseases. The copper levels in the human body are regulated by copper homeostasis, which maintains appropriate levels of copper in tissues and cells by controlling its absorption, transport, and storage. Cuproptosis is a distinct form of cell death ...

Deciphering mitochondrial dysfunction: Pathophysiological mechanisms in vascular cognitive impairment
Biomed Pharmacother. 2024 Apr 9;174:116428. doi: 10.1016/j.biopha.2024.116428. Online ahead of print.ABSTRACTVascular cognitive impairment (VCI) encompasses a range of cognitive deficits arising from vascular pathology. The pathophysiological mechanisms underlying VCI remain incompletely understood; however, chronic cerebral hypoperfusion (CCH) is widely acknowledged as a principal pathological contributor. Mitochondria, crucial for cellular energy production and intracellular signaling, can lead to numerous neurological impairments when dysfunctional. Recent evidence indicates that mitochondrial dysfunction-marked by oxid...

Co-administration of "L-Lysine, Vitamin C, and Zinc" increased the antioxidant activity, decreased insulin resistance, and improved lipid profile in streptozotocin-induced diabetic rats
CONCLUSIONS: The results of this study indicated that co-administration of Lys, a chemical chaperone, with two antioxidants (VC and Zn) potentiates its antidiabetic effects and prevent diabetic complications in rat model of diabetes.PMID:38599057 | DOI:10.1016/j.biopha.2024.116525 (Source: Biomedicine and pharmacotherapy = Biomedecine and pharmacotherapie)

In vivo effects of a selected thiourea derivative 1-(2-chlorobenzoyl)-3-(2,3-dichlorophenyl) against nociception, inflammation and gastric ulcerogenicity: Biochemical, histopathological and in silico approaches

Vitamin B < sub > 6 < /sub > alleviates chronic sleep deprivation-induced hippocampal ferroptosis through CBS/GSH/GPX4 pathway
Biomed Pharmacother. 2024 Apr 9;174:116547. doi: 10.1016/j.biopha.2024.116547. Online ahead of print.ABSTRACTSeveral studies have found that sleep deprivation (SD) can lead to neuronal ferroptosis and affect hippocampal function. However, there are currently no effective interventions. Vitamin B6 is a co-factor for key enzymes in the transsulfuration pathway which is critical for maintaining cell growth in the presence of cysteine deprivation. The results showed that SD inhibited cystine-glutamate antiporter light chain subunit xCT protein expression and caused cysteine deficiency, which reduced the synthesis of the glutat...
