Evaluation of lipid extraction methods for fatty acid quantification and compound-specific δ < sup > 13 < /sup > C and δ < sup > 2 < /sup > H < sub > n < /sub > analyses
In this study, we examined the efficacy of seven well-established sample preparation methods, specifically focusing on their effectiveness in total lipid and fatty acid extraction and their impact on compound-specific stable hydrogen (δ2H) and carbon (δ13C) isotope values. We also considered the repercussions of FA removal efficacy on residual bulk tissue δ2Hn analysis, because lipids typically have low δ2H values. Our findings showed that in most cases chloroform-based extraction methods outperformed those without chloroform. While discrepancies were not as evident for smaller organisms, such as plankton, marked varia...

Colony and bacterial LAMP: Simple alternative methods for bacterial determination
In this study, we developed colony and bacterial LAMP, which directly use bacterial colony and bacterial culture as the templates without DNA extraction for rapid and simple detection of bacteria. The end-point readouts were determined by naked eye under ultraviolet light, and real-time fluorescence curve was also used to confirm that the sensitivity of this method to Salmonella typhimurium and Bacillus cereus was 102 and 103 CFU/reaction, respectively. Results presented here provide alternative methods for colony and bacterial PCR that can greatly contribute to reliable and cost-effective diagnosis in resource-poor settin...

Comparison of the effect of Sodium Chloride concentration on protein determination: Bradford and Biuret methods
Anal Biochem. 2023 Dec 28:115450. doi: 10.1016/j.ab.2023.115450. Online ahead of print.ABSTRACTProtein quantification methods using spectrophotometry are widely used in laboratory routines for different purposes. Samples generally contain non-protein components that can interfere with the quality of the analysis. A simple and quick test with different concentrations of sodium chloride demonstrated that the Bradford method is significantly affected by the presence of salt, while Biuret remains stable. Therefore, the choice of method is an important factor in reducing errors and ensuring more reliable results.PMID:38159714 |...

Proficiently partitioning of bioactive peptide-ssDNA conjugates by microbead-assisted capillary electrophoresis (MACE)
Anal Biochem. 2023 Dec 27:115452. doi: 10.1016/j.ab.2023.115452. Online ahead of print.ABSTRACTLow-molecular drug discovery using DNA-encoded chemical library (DEL) is a powerful technology, although improving the partitioning efficiency of affinity ligands from DEL remains a challenge. Here, we assessed the usefulness of microbead-assisted capillary electrophoresis (MACE) for partitioning peptide-oligonucleotide conjugates (POCs), in which high selection pressure is applied because of different mobility of target-modified beads and POCs during CE. Despite their different charge characteristics, all POCs were well separate...

Development and application of LC-MS/MS method for the quantification of hydrogen sulfide in the eye
Anal Biochem. 2023 Dec 27:115448. doi: 10.1016/j.ab.2023.115448. Online ahead of print.ABSTRACTThere are limited studies that report the physiological levels of H2S in the eye. The currently available UV/Vis methods lack the required sensitivity and precision. Hence, the purpose of this study was to develop and validate a sensitive and robust pre-column derivatization LC-MS/MS method to measure changes in H2S levels in tissues from isolated porcine eyes. H2S was derivatized and an LC-MS/MS method was developed to monitor the derivatized product, Sulfide-dibimane (Sdb) using a reverse phase Waters Acquity BEH C18 column (1....

A simple and sensitive colorimetric approach for mecA gene analysis via exonuclease-III catalyzed signal cascade
Anal Biochem. 2023 Dec 27:115453. doi: 10.1016/j.ab.2023.115453. Online ahead of print.ABSTRACTAnalysis of mecA gene in Staphylococcus aureus (S. aureus) is essential for controlling infections in intensive care units (ICU) and preventing the use of ineffectual empirical treatments. However, quantitative determination of the mecA gene remains difficult. Herein, we propose a simple and sensitive colorimetric approach by integrating exonuclease-III (Exo-III) assisted signal cascade and G-quadruplex/hemin DNAzymes (G4 DNAzymes) catalyzed 2,2'-azino-bis (3-ethylben-zothiazoline-6-sulfonic acid) (ABTS) based color reaction. ITC analysis of polydisperse systems: Unravelling the impact of sample heterogeneity
Anal Biochem. 2023 Dec 24:115446. doi: 10.1016/j.ab.2023.115446. Online ahead of print.ABSTRACTBinding interactions often involve heterogeneous samples displaying a distribution of binding sites that vary in affinity and binding enthalpy. Examples include biological samples like proteins and chemically produced samples like modified cyclodextrins. Experimental studies often ignore sample heterogeneity and treat the system as an interaction of two homogeneous species, i.e. a chemically well-defined ligand binding to one type of site. The present study explores, by simulations and experiments, the impact of heterogeneity in ...
