SURE gel electrophoresis: A method for improved detection and purification of dilute nucleic acid samples
In this study a new approach is described for concentrating samples directly within gels called SURE (successive reloading) electrophoresis. The approach involves loading of dilute samples multiple times into a single well, with each loading followed by a brief pulse of electrical current before the next sample is loaded. The procedure generates single bands created by molecular stacking that exhibit strongly enhanced signal intensities and minimal band broadening. Using optimized voltages and time intervals as many as 20 successive loadings could be performed and up to 800 μL could be loaded into a single well. Gel extra...

LAMPrimers iQ: New primer design software for loop-mediated isothermal amplification (LAMP)
Anal Biochem. 2023 Nov 2:115376. doi: 10.1016/j.ab.2023.115376. Online ahead of print.ABSTRACTNucleic acids amplification is a widely used technique utilized for different manipulations with DNA and RNA. Although, polymerase chain reaction (PCR) remains the most popular amplification method, isothermal approaches are gained more attention last decades. Among these, loop-mediated isothermal amplification (LAMP) became an excellent alternative to PCR. LAMP requires an increased number of primers and, therefore, is considered a highly specific amplification reaction compared to PCR. LAMP primers design is still a non-trivial ...

Graphene oxide-enhanced colorimetric detection of Mec A gene based on toehold-mediated strand displacement
Anal Biochem. 2023 Oct 30:115365. doi: 10.1016/j.ab.2023.115365. Online ahead of print.ABSTRACTMec A, as a representative gene mediating resistance to β-lactam antibiotics in methicillin-resistant Staphylococcus aureus (MRSA), allows a new genetic analysis for the detection of MRSA. Here, a sensitive, prompt, and visual colorimetry is reported to detect the Mec A gene based on toehold-mediated strand displacement (TMSD) and the enrichment effect of graphene oxide (GO). The Mec A triggers to generate the profuse amount of signal units of single-stranded DNA (SG) composed of a long single-stranded base tail and a base head:...

Microarray fabrication techniques for multiplexed bioassay applications
Anal Biochem. 2023 Oct 30;683:115369. doi: 10.1016/j.ab.2023.115369. Online ahead of print.ABSTRACTMicroarrays are powerful tools for high-throughput bioassays that can extract information from tens of thousands of micro-spots consisting of biomolecules. This information is invaluable to many applications, such as drug discovery and disease diagnostics. Different applications of these microarrays need spots of different shapes, sizes, and chemistries to achieve their goals. Micro/nano-fabrication techniques are used to make microarrays with different feature structures and array densities for required assay procedures. Und...

Direct competitive assay for HER2 detection in human plasma using Bloch surface wave-based biosensors
Anal Biochem. 2023 Oct 30:115374. doi: 10.1016/j.ab.2023.115374. Online ahead of print.ABSTRACTThe overexpression and/or amplification of the HER2/neu oncogene has been proposed as a prognostic marker in breast cancer. The detection of the related peptide HER2 remains a grand challenge in cancer diagnosis and for therapeutic decision-making. Here, we used a biosensing device based on Bloch Surface Waves excited on a one-dimensional photonic crystal (1DPC) as valid alternative to standard techniques. The 1DPC was optimized to operate in the visible spectrum and the biosensor optics has been designed to combine label-free an...

Isoniazide modified Ag nanoparticles triggered photothermal immunoassay for carcinoembryonic antigen detection
Anal Biochem. 2023 Oct 25:115370. doi: 10.1016/j.ab.2023.115370. Online ahead of print.ABSTRACTAs the most well-known analytical tool, the thermometer has been extended to the field of biological analysis based on the photothermal effect. Herein, isoniazide modified Ag nanoparticles were prepared as nanolabels to build an immunoassay. The nanoparticles were characterized by transmission electron microscope (TEM), dynamic laser scattering (DLS), X-ray powder diffraction (XRD), and Fourier transform infrared (FT-IR). When the target protein was present, the sandwich immunoassay was developed and the photothermal reaction was...

A review on current diagnostic tools and potential optical absorption spectroscopy for HFMD detection
Anal Biochem. 2023 Oct 25:115368. doi: 10.1016/j.ab.2023.115368. Online ahead of print.ABSTRACTHand, Foot, and Mouth Disease (HFMD) is an outbreak infectious disease that can easily spread among children under the age of five. The most common causative agents of HFMD are enterovirus 71 (EV71) and coxsackievirus A16 (CVA16), but infection caused by EV71 is more associated with fatalities due to severe neurological disorders. The present diagnosis methods rely on physical examinations by the doctors and further confirmation by laboratories detection methods such as viral culture and polymerase chain reaction. Clinical signs ...
