GANSamples-ac4C: Enhancing ac4C site prediction via generative adversarial networks and transfer learning
This study introduces GANSamples-ac4C, a novel framework that synergizes transfer learning and generative adversarial network (GAN) to generate synthetic RNA sequences to train a better ac4C modification site prediction model. Comparative analysis reveals that GANSamples-ac4C outperforms existing state-of-the-art methods in identifying ac4C sites. Moreover, our result underscores the potential of synthetic data in mitigating the issue of data scarcity for biological sequence prediction tasks. Another major advantage of GANSamples-ac4C is its interpretable decision logic. Multi-faceted interpretability analyses detect key r...

COVID-19 electrochemical immunosensor with Ag-MOF: Rapid and high-selectivity nasal swab testing for effective detection
In this study, we leveraged the properties of synthesized Ag-MOF, including high porosity and increased flow intensity. Electrochemical techniques such as cyclic voltammetry (CV) and differential pulse voltammetry (DPV) were employed to develop an economical and portable sensor with exceptional selectivity for COVID-19 detection. The methodology involves the deposition of Ag-MOF onto the surface of a Glassy Carbon Electrode (GCE), which resulted in a progressive augmentation of electric current. Subsequently, the targeted antibodies were applied, and relevant tests were conducted. The sensor demonstrated the capacity to de...

An LC-MS/MS-based method for the simultaneous quantification of melatonin, cortisol and cortisone in saliva
Anal Biochem. 2024 Feb 29:115496. doi: 10.1016/j.ab.2024.115496. Online ahead of print.ABSTRACTDisturbances in the diurnal pattern are associated with several clinical and psychological conditions, including depression and fatigue. Salivary sampling for melatonin, cortisol and cortisone provides a non-invasive method for frequent sampling and obtaining biochemical insight into the diurnal pattern of individuals. Therefore, a new liquid chromatography-tandem mass spectrometry-based method for the measurement of salivary melatonin, cortisol and cortisone was developed and validated. The method required 250 μl saliva, used i...

CRISPR/Cas13a-triggered entropy-driven amplification for colorimetric and fluorescent dual-mode detection of microRNA
Anal Biochem. 2024 Feb 29:115499. doi: 10.1016/j.ab.2024.115499. Online ahead of print.ABSTRACTMicroRNAs (miRNAs) are crucial biomarkers for the early detection and monitoring of disease progression of chronic obstructive pulmonary disease (COPD). Herein, we have devised a method for detecting miRNA using a combination of colorimetric and graphene oxide-based fluorescent techniques. The target miRNA in our design could precisely activate the trans-cleavage activity of the CRISPR-Cas13a system. The activated Cas13a enzyme cuts the "rUrU" section in the P1 probe, generating a nicking site to induce entropy-driven amplificati...

Photophysical properties of DAPI in PVA films. Possibility of room temperature phosphorescence
Anal Biochem. 2024 Feb 28;689:115498. doi: 10.1016/j.ab.2024.115498. Online ahead of print.ABSTRACTWe studied the spectral properties of 4'-6-diamidino-2-phenylindole (DAPI) in poly (vinyl alcohol) (PVA) films. Absorption and fluorescence spectra, emission and excitation spectra, quantum yield, and fluorescence lifetime have been characterized. An efficient room temperature phosphorescence (RTP) of DAPI has been observed with UV and blue light excitations. A few hundred millisecond phosphorescence lifetime enables a gated detection with sufficient background reduction. We found the phosphorescent Quantum Yield of DAPI in P...

Time-resolved fluoroimmunoassay for Aspergillus detection based on anti-galactomannan monoclonal antibody from stable cell line
Anal Biochem. 2024 Feb 23;689:115494. doi: 10.1016/j.ab.2024.115494. Online ahead of print.ABSTRACTInvasive Aspergillosis is a high-risk illness with a high death rate in immunocompromised people due to a lack of early detection and timely treatment. Based on immunology study, we achieved an efficient production of anti-galactomannan antibody by Chinese hamster ovary (CHO) cells and applied it to time-resolved fluoroimmunoassay for Aspergillus galactomannan detection. We first introduced dual promoter expression vector into CHO host cells, and then applied a two-step screening strategy to screen the stable cell line by met...
