Design and construction of Carboxylesterase 2c Gene knockout Rats by CRISPR/Cas9
CONCLUSION: The Ces2c KO rat model was successfully constructed by CRISPR/Cas9 system. This rat model can not only be used as an important tool to study the drug metabolism mediated by Ces2, but also as an important animal model to study the physiological function of Ces2.PMID:36694315 | DOI:10.2174/1389200224666230123140919 (Source: Current Drug Metabolism)

Clinical outcomes of Medical management options for chronic anal fissures in a long term follow up: systematic review and meta-analysis
Conclusions: Calcium channel blockers performed well regarding the healing of CAF when compared to others in long-term follow-up. The superiority of botulinum toxin was not evidenced compared to topical treatments. More studies are needed to better assess recurrence rates. (Source: Digestive Diseases)

Comparing the efficacy and safety of different analgesic strategies after open hemorrhoidectomy: a systematic review and network meta-analysis
ConclusionDiltiazem ointment may be the most effective medication for pain relief following open hemorrhoidectomy, and it can dramatically reduce pain within one week of surgery. The second and third recommended medications are BT and sucralfate ointment. GTN has a significant advantage in alleviating pain 24  h after open hemorrhoidectomy, but whether it causes headache is debatable; thus, it should be used with caution. EA’s analgesic efficacy is still unknown. There was limited evidence on the safety of the intervention in this study, and it was simply presented statistically. Online ahead of print.ABSTRACTOBJECTIVE: To determine if left ventricular systolic function on echocardiography, systemic blood pressure, and electrocardiography change with a clinically accepted intravenous (IV) diltiazem constant rate infusion (CRI) compared to a control.ANIMALS: 10 healthy client-owned adult dogs.PROCEDURES: Prospective, masked, crossover study from May 27, 2021, to August 22, 2021. Dogs were randomized to receive diltiazem (loading dose of 240 μg/kg, IV followed by a CRI of 6 μg/kg/min for 300 minutes) or the same volume of 5% dextrose in w...

Repetitive multiform narrow complex tachycardia
A 73-year-old man presented to the emergency department with palpitations. After experiencing transient exertional palpitations and dyspnea a few months earlier, investigation revealed moderate left ventricular dysfunction and extensive coronary artery disease with evidence of prior inferior infarction. He underwent bypass grafting and remained symptom free with normalization of his ejection fraction until the current presentation. Figure  1 demonstrates his presenting rhythm that did not respond to adenosine but terminated with intravenous diltiazem. (Source: Heart Rhythm)

Can Targeting Sphincter Spasm Reduce Post-Haemorrhoidectomy Pain? A Systematic Review and Meta-Analysis
ConclusionEvidence suggests that lateral sphincterotomy, administration of botulinum toxin and the application of topical diltiazem or GTN can reduce post-operative pain after haemorrhoidectomy. Lateral sphincterotomy should not be routinely used due to the risk of incontinence. (Source: World Journal of Surgery)

Glucocorticoid Effects on Working Memory Impairment Require L-Type Calcium Channel Activity Within Prefrontal Cortex
Neurobiol Learn Mem. 2022 Nov 18:107700. doi: 10.1016/j.nlm.2022.107700. Online ahead of print.ABSTRACTPrevious findings have indicated that glucocorticoid hormones impair working memory via an interaction with the β-adrenoceptor-cAMP signaling cascade to rapidly increase cAMP-dependent protein kinase (PKA) activity within the prefrontal cortex (PFC). However, it remains elusive how such activation of PKA can affect downstream cellular mechanisms in regulating PFC cognitive function. PKA is known to activate L-type voltage-gated Ca2+ channels (LTCCs) which regulate a broad range of cellular processes, including neuronal e...
