Dabrafenib and trametinib in Langerhans cell histiocytosis and other histiocytic disorders
Haematologica. 2023 Sep 21. doi: 10.3324/haematol.2023.283295. Online ahead of print.ABSTRACTStandard treatment for Langerhans Cell Histiocytosis (LCH) is chemotherapy, with high failure rates. Since MAP-kinase activating mutations are found in most cases, BRAF- and MEKinhibitors have been used successfully to treat patients with refractory or relapsed disease. However, data on long-term responses in children are limited and no data exist on their use as first-line therapy. We treated thirty-four patients (26 LCH, 2 Juvenile Xanthogranuloma, 2 Rosai-Dorfman Disease, 4 presumed single site-CNS histiocytosis) with either dab...

Racism behind the screen: Examining the mediating and moderating relationships between anonymity, online disinhibition, and cyber-racism
J Pers Soc Psychol. 2023 Sep 21. doi: 10.1037/pspa0000360. Online ahead of print.ABSTRACTCyber-racism has emerged as a societal issue that affects many youths and adults; however, no published work has elucidated the psychological processes germane to predicting cyber-racism perpetration. Theory-without data to support its postulates-argues that online disinhibition mediates the relationship between anonymity afforded the online user and cyber-racism. The purpose of the current research was to examine this prediction and add to the theory by testing additional mediators and moderators. Six empirical studies tested this the...

Phenolic Substitution in Fidaxomicin: A Semisynthetic Approach to Antibiotic Activity Across Species
Chembiochem. 2023 Sep 20:e202300570. doi: 10.1002/cbic.202300570. Online ahead of print.ABSTRACTFidaxomicin (Fdx) is a natural product antibiotic with potent activity against Clostridioides difficile and other Gram-positive bacteria such as Mycobacterium tuberculosis. Only few Fdx derivatives have been synthesized and examined for their biological activity in the 50 years since its discovery. Fdx has a well-studied mechanism of action, namely inhibition of the bacterial RNA polymerase. Yet, the targeted organisms harbor different target protein sequences, which poses a challenge for the rational development of new semisynt...

Prospective associations of neighborhood healthy food access and walkability with weight status in a regional pediatric health system
CONCLUSIONS: Findings provide evidence that residing in a neighborhood with healthy food access and walkability may support a healthy weight trajectory in children. Findings on changes in the neighborhood environment suggested that improved walkability in the neighborhood may support children's healthy weight. The greater and more consistent findings among movers may be due to movers experiencing greater changes in neighborhood features than the changes that typically occur within a neighborhood over a short period of time. Future research is needed to investigate more robust environmental changes to neighborhoods.PMID:377...

Distinct Features of Plasma Ultrashort Single-Stranded Cell-Free DNA as Biomarkers for Lung Cancer Detection
CONCLUSIONS: These observations indicate that the distinct biological processes of uscfDNA and that FE peaks, fragmentomics, end-motifs, and G-Quad signatures are uscfDNA features with promising biomarker potential. These findings further justify its exploration as a distinct class of biomarker to augment pre-existing liquid biopsy approaches.PMID:37725931 | DOI:10.1093/clinchem/hvad131

Distinctive Arbutin-Containing Markers: Chemotaxonomic Significance and Insights Into the Evolution of Proteaceae Phytochemistry
Chem Biodivers. 2023 Sep 19:e202301112. doi: 10.1002/cbdv.202301112. Online ahead of print.ABSTRACTNatural products isolation studies of eight endemic Tasmanian Proteaceae species - Agastachys odorata, Persoonia juniperina, Hakea megadenia, Hakea epiglottis, Orites diversifolius, Orites acicularis,Orites revolutus, and Telopea truncata - and three endemic Australian Proteaceae species Banksia serrata, Banksia praemorsa, and Banksia marginata were undertaken. Two previously unreported glycoside-derived natural products were identified, in addition to four other tremendously rare arbutin esters. The results of this study prov...

Uncovering associations between gender nonconformity, psychosocial factors, and mental health in adolescents: a birth cohort study
CONCLUSIONS: Persistent gender nonconformity during adolescence is associated with subsequent mental health. Psychosocial factors play a vital mediating role in this association, highlighting the essential need for social intervention and change to reduce stigmatization and ameliorate mental health challenges.PMID:37721216 | DOI:10.1017/S0033291723002623
