Osteogenic Potential of the Transcription Factor c-MYB
In conclusion, c-Myb plays a role in the osteogenesis of long bones by inducing osteogenic genes and causing the enhancement of mineral matrix production. This action of the transcription factor c-Myb might be of interest in the future for the establishment of novel approaches to tissue regeneration.

Effect of RNA Interference-Mediated Suppression of p75 on the Viability of Rat Notochordal Cells.
CONCLUSIONS: siRNA-mediated suppression of p75 inhibited apoptosis and increased proliferation of notochordal cells under conditions of serum deprivation, suggesting that RNAi might serve as a novel therapeutic approach for disc degeneration caused by insufficient viability of disc cells through the suppression of the expression of harmful genes. HIF-1-Dependent IL-6 Activation in Articular Chondrocytes Initiating Synovitis in Femoral Head Ischemic Osteonecrosis
Conclusions: Induction of ischemic osteonecrosis results in IL-6 production in the articular cartilage through an HIF-1-dependent pathway. IL-6 produced by hypoxic articular chondrocytes stimulates inflammatory cytokine responses in synovial cells, which were significantly decreased by tocilizumab. Clinical Relevance: This study provides new insight into the inherent relationship between the induction of ischemia and the initiation of hip synovitis following ischemic osteonecrosis and suggests a potential therapeutic target in the treatment of the synovitis.

Sonic Hedgehog Promotes Cementoblastic Differentiation via Activating the BMP Pathways
Abstract Although sonic hedgehog (SHH), an essential molecule in embryogenesis and organogenesis, stimulates proliferation of human periodontal ligament (PDL) stem cells, the effects of recombinant human SHH (rh-SHH) on osteoblastic differentiation are unclear. To reveal the role of SHH in periodontal regeneration, expression of SHH in mouse periodontal tissues and its effects on the osteoblastic/cementoblastic differentiation in human cementoblasts were investigated. SHH is immunolocalized to differentiating cementoblasts, PDL cells, and osteoblasts of the developing mouse periodontium. Addition of rh-SHH increas...

Regulation of BMP2‐induced intracellular calcium increases in osteoblasts
In this study, intracellular Ca2+ concentration ([Ca2+]i) upon BMP2 application, focal adhesion kinase (FAK) and Src activities were measured in the MC3T3‐E1 osteoblast cell line using fluorescence resonance energy transfer‐based biosensors. Increase in [Ca2+]i, FAK, and Src activities were observed during BMP2 stimulation. The removal of extracellular calcium, the application of membrane channel inhibitors streptomycin or nifedipine, the FAK inhibitor PF‐573228 (PF228), and the alkaline phosphatase (ALP) siRNA all blocked the BMP2‐stimulated [Ca2+]i increase, while the Src inhibitor PP1 did not. In contrast, a gen...

Heterozygous PTCH1 Mutations Impact the Bone Metabolism in Patients with Nevoid Basal Cell Carcinoma Syndrome Likely by Regulating SPARC Expression
This study sought to investigate the mechanism underlying heterozygous PTCH1 mutation‐mediated abnormal bone metabolism in patients with NBCCS. Stromal cells were isolated from the fibrous capsules of patients with NBCCS‐associated or non‐syndromic keratocystic odontogenic tumors and non‐syndromic tumor stromal cells without PTCH1 mutations served as controls. Germline PTCH1 heterozygous mutations were confirmed in all NBCCS samples and differential protein expression was identified using tandem mass tag‐labeled proteomics analysis. Our findings revealed that osteonectin/SPARC expression was significantly downreg...

Efficient, Low-Cost Nucleofection of Passaged Chondrocytes
This study identifies a cost-effective, efficient method of nonviral nucleofection of bovine-passaged chondrocytes using known buffer formulations. Human-passaged chondrocytes could also be successfully nucleofected in 1M buffer. Thus this method should facilitate cost-efficient gene targeting of cells used for articular cartilage repair in a research setting.
