A Randomized Trial of Penicillin Skin Testing Versus 2-Step Direct Challenge for Penicillin Allergy During Pregnancy
Penicillins are first-line therapy for peripartum prophylaxis in Group B streptococcal-colonized women, making delabeling of PCN allergy in this population particularly important. There is scant data regarding the safety of direct challenges in penicillin-allergic pregnant women.

PARP-14 Promotes Survival of Mammalian α but Not β Pancreatic Cells Following Cytokine Treatment
Conclusion This study must be seen as a first piece of a puzzle where PARP-14, JNKs and PJ-34 play key roles in the pancreatic microenvironment and provide starting points from which to explore further. Author Contributions VS-P conceived the project and designed the experiments together with MP and CD. MR and FD performed molecular experiments as RT-PCR and western analysis. CS and MS performed confocal analysis. FD, NM, and VB performed cytofluorimetric analysis. FD and MC were in charge of cell culture and treatment. FD performed computational and statistical data analysis, together with AT-S. VS-P and FD wrote the p...

Geniposide Alleviates Glucocorticoid-Induced Inhibition of Osteogenic Differentiation in MC3T3-E1 Cells by ERK Pathway
Conclusion In summary, we demonstrated that geniposide alleviated GC-induced osteogenic suppression in MC3T3-E1 cells. The effects of geniposide were at least partially associated with activating ERK signaling pathway via GLP-1 receptor. Geniposide might be a potential therapeutic agent for protection against GC-induced osteoporosis. Author Contributions BX, DX, CZ, and LW participated in research design. BX, JW, YL, XW, and ZZ conducted the experiments. BX, DX, and LW contributed new reagents or analytic tools and wrote or contributed to the writing of the manuscript. BX, JW, CZ, and DX performed the data analysis. Fu...

Focused Ultrasound Improves NK-92MI Cells Infiltration Into Tumors
Conclusion In conclusion, we demonstrated that the combination of FUS and microbubbles could increase the NK-92MI cells’ infiltration into tumors in this study. The combination of FUS and NK-92MI had a much better therapeutic effect when compared with the PBS group, but no superior therapeutic effect when compared with the NK-92MI group. In total, FUS and microbubbles can improve NK cells’ infiltration into tumors, but future work is still needed to improve NK-92MI cells’ delivery efficiency for solid tumor treatment. Ethics Statement This study was carried out in accordance with the recommendations...

Direct oral amoxicillin challenge without preliminary skin testing in patients with reported penicillin allergy
10% of hospitalized patients report penicillin allergy; studies indicate that ∼98% are not truly allergic. Unconfirmed penicillin allergy labels pose public health risks, and evaluation is recommended to improve antibiotic stewardship. While the most widely accepted protocol is penicillin skin testing (PST) followed by oral amoxicillin challenge, time constraints and resour ces may preclude this. Recent literature supports the safety and efficacy of direct oral amoxicillin challenge in low-risk individuals.

Quality assessment in general practice: diagnosis and antibiotic treatment of acute respiratory tract infections.
CONCLUSION: The findings indicate an overuse of antibiotics for RTIs in Danish general practice. Especially management of acute rhinosinusitis and acute bronchitis should be targeted in future quality improvement projects. KEY POINTS: To improve antibiotic prescribing in general practice it is important to focus on both the diagnostic process and the prescribing patterns. The findings indicate an overuse of antibiotics for acute respiratory tract infections in Danish general practice. Especially the diagnostic process and antibiotic prescribing patterns for acute rhinosinusitis and acute bronchitis could benefit from ...

Does this child really have a penicillin allergy?
Abstract Penicillins, the most prescribed paediatric medications worldwide, are also the most commonly reported cause of medication allergy, although this is rarely confirmed. An oral penicillin challenge is considered the gold standard in assessing children with suspected allergy but is seldom performed due to lack of appropriately trained staff and insufficient facilities. We introduced a standardised nurse-led protocol to evaluate children with suspected penicillin allergy fulfilling low risk criteria. In total, 40 children participated, including 22 girls and 18 boys, of which 38 met study criteria. There were...
