Effects of empagliflozin on collagen biomarkers in patients with heart failure: Findings from the EMPEROR trials
ConclusionOur observations are consistent with experimental observations that empagliflozin down-regulates profibrotic signalling. The importance of such an effect for the clinical benefits of SGLT2i in HF remains to be elucidated. (Source: European Journal of Heart Failure)

Empagliflozin Alleviates Diabetes-Induced Cognitive Impairments by Lowering Nicotinamide Adenine Dinucleotide Phosphate Oxidase-4 Expression and Potentiating the Antioxidant Defense System in Brain Tissue of Diabetic Rats
CONCLUSION: Uncontrolled diabetes negatively impacts mental function and impairs learning and cognitive performance via oxidative stress induction, the Nox-4 enzyme playing a central role. Empagliflozin reverses these effects, improving cognitive ability via promoting the anti-oxidative system and damping Nox-4 free radical generator enzyme expression. Therefore, empagliflozin is a promising treatment, providing both antidiabetic and extra-glycemic benefits for improving brain function in the diabetic milieu.PMID:38141785 | DOI:10.1016/j.bbr.2023.114830 (Source: Brain Research)

Identification of Risk Factors Associated with Acute Kidney Injury in Patients Taking Sodium ‐Glucose Cotransporter‐2 Inhibitors
ConclusionsThe transient eGFR decline with SGLT2 inhibitor initiation should be expected and is generally not an indication to discontinue therapy. Future initiatives should be directed at increasing knowledge of monitoring recommendations for these agents. (Source: Pharmacotherapy: The Journal of Human Pharmacology and Drug Therapy)

Comparative cardiovascular benefits of individual SGLT2 inhibitors in type 2 diabetes and heart failure: a systematic review and network meta-analysis of randomized controlled trials
ConclusionSGLT2is significantly reduce the composite CV death/HFH outcome. Among them, canagliflozin may be considered the preferred treatment for patients with diabetes and a history of heart failure, but it may also be associated with an increased risk of any adverse events compared to other SGLT2is. However, a sensitivity analysis focusing on HF-specific trials identified sotagliflozin as the most likely agent to reduce CV death/HFH, followed by empagliflozin and dapagliflozin.Systematic review registrationhttps://www.crd.york.ac.uk/prospero/, identifier CRD42022353754. Assessing early tubular protective effects of SGLT2 inhibitor empagliflozin against type 2 diabetes mellitus using functional magnetic resonance imaging
ConclusionsSGLT2 inhibitor empagliflozin can reduce UACR and eGFR levels in early type 2 diabetic patients with normal proteinuria. Moreover, empagliflozin therapy led to an increase in the MD value and a decrease in the MR2* value of the renal medulla, evidencing the early tubular protective effects of this therapy. Empagliflozin treatment rescues abnormally reduced Na currents in ventricular cardiomyocytes from dystrophin-deficient mdx mice
Am J Physiol Heart Circ Physiol. 2023 Dec 15. doi: 10.1152/ajpheart.00729.2023. Online ahead of print.ABSTRACTCardiac arrhythmias significantly contribute to mortality in Duchenne muscular dystrophy (DMD), a severe muscle illness caused by mutations in the gene encoding for the intracellular protein dystrophin. A major source for arrhythmia vulnerability in patients with DMD is impaired ventricular impulse conduction, which predisposes for ventricular asynchrony, decreased cardiac output and the development of reentrant circuits. Using the dystrophin-deficient mdx mouse model for human DMD, we previously reported that the ...
