The GLP-1-mediated Gut-Kidney Cross Talk in Humans: Mechanistic Insight
Am J Physiol Cell Physiol. 2023 Dec 18. doi: 10.1152/ajpcell.00476.2023. Online ahead of print.ABSTRACTIncretin-based therapy is an anti-diabetic and anti-obesity approach mimicking glucagon-like peptide 1 (GLP-1) with additional end-organ protection. This review solely focuses on randomized, controlled mechanistic human studies on renal effect of GLP-1. There is not consensus about the localization of GLP-1 receptors (GLP-1R) in human kidneys. Rodent and primate data suggest GLP-1R distribution in smooth muscle cells in the preglomerular vasculature. Native GLP-1 and GLP-1R agonists elicit renal effects. Independently of ...

Altered glucagon and GLP-1 responses to oral glucose in children and adolescents with obesity and insulin resistance
CONCLUSIONS: The OIR-group had elevated fasting concentrations of glucagon and GLP-1, and higher glucagon, but lower GLP-1 responses during an OGTT compared to the OIS- and NW-groups. In contrast, the OIS-group had similar hormone responses to the NW-group.PMID:38087928 | DOI:10.1210/clinem/dgad728 (Source: The Journal of Clinical Endocrinology and Metabolism)

An Extended Minimal Model of OGTT: Estimation of Alpha- and Beta-Cell Dysfunction, Insulin Resistance, and the Incretin Effect
Am J Physiol Endocrinol Metab. 2023 Dec 13. doi: 10.1152/ajpendo.00278.2023. Online ahead of print.ABSTRACTLoss of insulin sensitivity, alpha- and beta-cell dysfunction, and impairment in incretin effect have all been implicated in the pathophysiology of type 2 diabetes (T2D). Parsimonious mathematical models are useful in quantifying parameters related to the pathophysiology of T2D. Here we extend the minimum model developed to describe the glucose-insulin-glucagon dynamics in the intravenous glucose infusion (IIGI) experiment to the oral glucose tolerance test (OGTT). The extended model describes glucose and hormone dyna...
