Advantage of ostarine over raloxifene and their combined treatments for muscle of estrogen-deficient rats
ConclusionOST had beneficial effects on muscle in OVX rats. Side effects of OST on the uterus and serum electrolytes should be considered before using it for therapeutic purposes. RAL and RAL  + OST had less effect on muscle and showed endocrinological side effects on pituitary–gonadal axis. (Source: Journal of Endocrinological Investigation)

Comparative Pharmacokinetic Profiles of a Novel Low ‐Dose Micronized Formulation of Raloxifene 45 mg (AD‐101) and the Conventional Raloxifene 60 mg in Healthy Subjects
This study was designed as an open-label, randomized, 2-treatment-period, crossover study with a 2-week washout period. Two treatments consisted of micronized raloxifene 45  mg daily; and conventional raloxifene 60 mg daily administered in fasting conditions. Plasma raloxifene concentrations were determined by a validated method using ultra-fast liquid chromatography–tandem mass spectrometry, and pharmacokinetic parameters were calculated using a noncompartmental m odel. In total, 49 subjects completed the study. The geometric mean ratio (micronized/conventional) of the maximum concentration and the area under the plas...

Hormone therapy for sexual function in perimenopausal and postmenopausal women
CONCLUSIONS: Hormone therapy treatment with estrogen alone probably slightly improves the sexual function composite score in women with menopausal symptoms or in early postmenopause (within five years of amenorrhoea), and in unselected postmenopausal women, especially in the lubrication, pain, and satisfaction domains. We are uncertain whether estrogen combined with progestogens improves the sexual function composite score in unselected postmenopausal women. Evidence regarding other hormone therapies (synthetic steroids and SERMs) is of very low quality and we are uncertain of their effect on sexual function. The current e...

Synthesis of raloxifene-like quinoxaline derivatives by intramolecular electrophilic cyclization with disulfides
Bioorg Med Chem Lett. 2023 Jul 31:129415. doi: 10.1016/j.bmcl.2023.129415. Online ahead of print.ABSTRACTThe intramolecular electrophilic cyclization of alkynes with disulfides to form thieno[2,3-b]quinoxaline structures and to introduce thioether substituents afforded quinoxaline derivatives (7a-7d, 8a-8d). Among obtained eight derivatives, the raloxifene analogues (7c, 8b) showed specifically high cytotoxicity against breast cancer cells (SK-BR-3), and raloxifene analogues (8a) showed the highest cytotoxicity against human leukemia cells (HL-60). None of the raloxifene analogues (7a-7d, 8a-8d) showed cytotoxicity against...
