Multipotent stem cell-derived retinal ganglion cells in 3D culture as tools for neurotrophic factor gene delivery system development

We report the development of an RGC cell model, XFC-3 cells, as a tool for preclinical screening of three non-viral gene delivery systems (made from dicationic gemini surfactants, single-walled carbon nanotubes, and K2-lipopolyamine polymers) for glaucoma neurotrophic factor gene therapy. XFC-3 cells were isolated from differentiated MRSCs and characterized by gene expression profiling and neurite extension activity. XFC-3 cells also demonstrated effectiveness and versatility in 3D co-culture bioassay models for therapeutic protein bioactivity assessment.
