Free docosahexaenoic acid promotes ferroptotic cell death via lipoxygenase dependent and independent pathways in cancer cells
ConclusionDHA can effectively promote ferroptosis-mediated tumor killing by increasing intracellular lipid peroxidation. Both ALOX5 dependent and independent pathways are involved in DHA-FIN induced ferroptosis. And during this process, free DHA plays an important role.

Taurine stimulates protein synthesis and proliferation of C2C12 myoblast cells through the PI3K-ARID4B-mTOR pathway
In this study, we investigated the role and underlying molecular mechanism of Tau on protein synthesis and proliferation of C2C12 myoblast cells. Cells were treated with Tau (0, 60, 120, 180 and 240 μM) for 24 h. Tau dose-dependently promoted protein synthesis, cell proliferation, mTOR phosphorylation, and also AT-rich interaction domain 4B (ARID4B) expression, with the best stimulatory effects at 120 μM. LY 294002 treatment showed that Tau promoted ARID4B expression in a PI3K-dependent manner. ARID4B knockdown (by siRNA transfection for 24 h) prevented Tau from stimulating protein synthesis and cell proliferation, where...

The BMSC-derived exosomal lncRNA Mir9-3hg suppresses cardiomyocyte ferroptosis in ischemia-reperfusion mice via the Pum2/PRDX6 axis
The exosomal long noncoding RNAs (lncRNAs) have been reported to have cardioprotective effects on ischemia-reperfusion (I/R) injury by hindering ferroptosis, but the role of lncRNA Mir9-3 host gene (Mir9-3hg) in cardiac I/R injury remains unclear. Exosomes were extracted from mouse bone marrow mesenchymal stem cells (BMSCs) and identified by detecting the exosome specific marker levels, and the results showed that Mir9-3hg was highly expressed in BMSCs-Exo. Hypoxia/reoxygenation (H/R)-treated HL-1 mouse cardiomyocytes were incubated with exosomes extracted from BMSCs transfected with Mir9-3hg siRNA.

Homocysteine-induced decrease in HUVEC cells ’ resistance to oxidative stress is mediated by Akt-dependent changes in iron metabolism
ConclusionsThese data indicate that Hcy induces an increase in cellular ferritin level, and the process is mediated by alterations in Akt-FOXO3a signaling pathway.

Walnut polyphenol extracts inhibit Helicobacter pylori-induced STAT3Tyr705 phosphorylation through activation of PPAR- γ and SOCS1 induction.
Walnut polyphenol extracts inhibit Helicobacter pylori-induced STAT3Tyr705 phosphorylation through activation of PPAR-γ and SOCS1 induction. J Clin Biochem Nutr. 2020 Nov;67(3):248-256 Authors: Park JM, An JM, Han YM, Surh YJ, Hwang SJ, Kim SJ, Hahm KB Abstract The health beneficial effects of walnut plentiful of n-3 polyunsaturated fatty acid had been attributed to its anti-inflammatory and anti-oxidative properties against various clinical diseases. Since we have published Fat-1 transgenic mice overexpressing 3-desaturase significantly mitigated Helicobacter pylori (H. pylori)-associated gastric pat...

Purple sweet potato color protects against hepatocyte apoptosis through Sirt1 activation in high-fat-diet-treated mice.
Conclusion: PSPC protected against HFD-induced hepatic apoptosis by promoting Sirt1- dependent inhibition of p53-apoptotic pathway and facilitation of Akt survival pathway. This study indicates that PSPC is a candidate for nutritional intervention of NAFLD. PMID: 32110174 [PubMed - as supplied by publisher]

Amentoflavone inhibits tumorsphere formation by regulating the Hedgehog/Gli1 signaling pathway in SUM159 breast cancer stem cells
This study aimed to investigate the effects of amentoflavone, a natural compound identified from Chamaecyparis obtusa, on stemness of basal-like SUM159 breast CSCs, and its underlying mechanism of action. We found that amentoflavone suppressed tumorsphere formation and regulated the expression of the stem cell markers CD44, CD24, and ALDH1 in a dose dependent manner, and inhibited Hedgehog (Hh) signaling by down-regulating Smo, Gli1, OCT4, and NANOG. Knock-down of Gli1 by siRNA significantly affected the expression of CD44, CD24, ALDH1, OCT4, and NANOG, and reduced tumorsphere formation efficiency in SUM159 breast CSCs. Ou...

Flavonoids from Livistona chinensis fruit ameliorates LPS/D-GalN-induced acute liver injury by inhibiting oxidative stress and inflammation
This study investigated the hepatoprotective effect and mechanism of flavonoids from Livistona chinensis fruit (FLCF). Mice received FLCF pre-treatment for 7 days and induced liver injury with LPS+D-GalN. RAW264.7 cells were triggered inflammation by LPS and received FLCF for 24 h. The result showed that FLCF ameliorated the liver pathology in mice, decreased the levels of AST, ALT, MDA, but increased SOD and GSH. Moreover, FLCF inhibited TLR4/NF-κB signaling, while activated Nrf2/HO-1 signaling in LPS+D-GalN mice. These effects were consistent with the in vitro data revealing that FLCF suppressed TLR4/NF-κB signalin...

Induction of sestrin 2 is associated with fisetin-mediated apoptosis in human head and neck cancer cell lines.
In this study, the influence of fisetin on the growth and apoptosis of HNCCs were examined. In HSC3 cells, fisetin treatment reduced the viability and induced apoptosis. Through the results from the screening of the expression profile of apoptosis-related genes, sestrin 2 (SESN2) was functionally involved in fisetin-mediated apoptosis showing the knockdown of SESN2 by siRNA clearly restored fisetin-induced apoptosis. In addition, fisetin reduced the protein expression levels of phospho-mTOR (p-mTOR) and Mcl-1, which are the downstream molecules of SESN2. It also induced PARP cleavage by inducing an increase in the expressi...

Modulation of GSK - 3β/β - catenin cascade by commensal bifidobateria plays an important role for the inhibition of metaflammation-related biomarkers in response to LPS or non-physiological concentrations of fructose: An in vitro study
ConclusionWe describe a regulatory mechanism in which commensal bifidobacteria prevent LPS- and/or fructose-mediated transcriptional activation of metabolic disorder-causing factors (IL-1β, IL-6, TNF-α) and disruption of normal cellular energy metabolism by attenuating NF-kB activity and interfering with the Wnt/β-catenin pathway. The latter positively regulates NF-κB activity, thereby simultaneously reducing cellular triglyceride concentrations and enhancing cellular glucose consumption. This may be a molecular basis by which commensal bifidobacteria enhance intrinsic cellular tolerance against excess consumption of e...

Commensal lactic acid-producing bacteria affect host cellular lipid metabolism through various cellular metabolic pathways: Role of mTOR, FOXO1, and autophagy machinery system
ConclusionLAPB prevent high- fat diet/endotoxin/inflammation-mediated disruption of normal cellular lipid metabolism by inhibiting mTOR/FOXO1/NF-kB activity and enhancing the occurrence of autophagy. This may be a molecular basis by which LAPB enhance intrinsic cellular tolerance against excess calorie consumption and participate in homeostatic regulation of metabolic processes in vivo.Graphical abstract
