PARP1-siRNA suppresses human prostate cancer cell growth and progression.
Authors: Lai Y, Kong Z, Zeng T, Xu S, Duan X, Li S, Cai C, Zhao Z, Wu W Abstract Poly (ADP-ribose) polymerase (PARP) inhibitors, such as olaparib or rucaparib, have shown treatment efficacy in BRCA1/2-deficient tumors. However, since PARP inhibitors (PARPi) mainly modulate the activation of PARP but not its expression, whether small interfering RNA (siRNA) specific to PARP has the same function as PARPi has not been well defined. In the present study it was demonstrated that PARP1-siRNA could reduce prostate cancer (PCa) cell progression regardless of the BRCA1/2 mutation. PARP1 silencing could significantly inhibi...

Cooperative Treatment of Metastatic Breast Cancer Using Host–Guest Nanoplatform Coloaded with Docetaxel and siRNA
Conventional chemotherapy shows moderate efficiency against metastatic cancer since it targets only part of the mechanisms regulating tumor growth and metastasis. Here, gold nanorod (GNR)‐based host‐guest nanoplatforms loaded with docetaxel (DTX) and small interfering RNA (siRNA)‐p65 (referred to as DTX‐loaded GNR (GDTX)/p65) for chemo‐, RNA interference (RNAi), and photothermal ablation (PTA) cooperative treatment of metastatic breast cancer are reported. To prepare the nanoplatform, GNRs are first coated with cyclodextrin (CD)‐grafted polyethylenimine (PEI) and then loaded with DTX and siRNA through host–gu...

Folate appended cyclodextrins for drug, DNA, and siRNA delivery.
Abstract Drug, DNA, and siRNA delivery systems based on cyclodextrin (CD) core and connected with folate (FA) via various linkers are presented. They include simple mono-derivatized cyclodextrins as well as cyclodextrins with higher degree of substitution, both in their primary and secondary sides. Examples of simple polymers and dendrimers are also discussed. Such carriers possess properties inherent to both of their components. Cyclodextrin provides the ability to encapsulate organic molecules in its inner cavity, thus improving their solubility in water, bioavailability, and stability, while FA assures targetin...

Abstract 3948: Identification of a new therapeutic target in prostate cancer from siRNA screening in Docetaxel-resistant cells
Conclusion: We identified a new chaperone harbouring an enzymatic activity which could be a relevant therapeutic target in chemoresistant CRPC. We are now focusing on the identification of a specific inhibitor in order to validate the role of this target in two Docetaxel-resistant prostate cancer mice models established within the laboratory.Citation Format: Marine Garrido, Nicolas J-p Martin, Catherine Gaudin, Elaine Del Nery, Jacques Camonis, Franck Perez, Stéphanie Lerondel, Alain Le Pape, Karim Fizazi, Anne Chauchereau. Folate-targeted star-shaped cationic copolymer co-delivering docetaxel and MMP-9 siRNA for nasopharyngeal carcinoma therapy.
Authors: Liu T, Wu X, Wang Y, Zhang T, Wu T, Liu F, Wang W, Jiang G, Xie M Abstract The co-delivery of drug and gene has become the primary strategy in cancer therapy. Based on our previous work, to co-deliver docetaxel (DOC) and MMP-9 siRNA more efficiently for HNE-1 nasopharyngeal carcinoma therapy, a folate-modified star-shaped copolymer (FA-CD-PLLD) consisting of β-cyclodextrin (CD) and poly(L-lysine) dendron (PLLD) was synthesized, and then used for DOC and MMP-9 co-delivery. Different from commonly used amphiphilic copolymers micelles, the obtained CD derivative could be used directly for the combinatorial d...

Targeted Nanoparticle for Co ‐delivery of HER2 siRNA and a Taxane to Mirror the Standard Treatment of HER2+ Breast Cancer: Efficacy in Breast Tumor and Brain Metastasis
This research paper describes a new therapeutic candidate based on a nanoparticle that co-delivers siRNA against human epidermal growth factor receptor type 2 (HER2), trastuzumab, and docetaxel to treat advanced HER2+ breast cancer. The nanotherapeutic is more effective and safer than the free drug counterparts. It inhibits drug-resistant orthotopic and brain-metastatic HER2+ breast tumors in mice. AbstractThe first-line treatment of advanced and metastatic human epidermal growth factor receptor type 2 (HER2+) breast cancer requires two HER2-targeting antibodies (trastuzumab and pertuzumab) and a taxane (docetaxel or pacli...

CD-PLLD co-delivering docetaxel and MMP-9 siRNA plasmid for nasopharyngeal carcinoma therapy in  vivo.
CD-PLLD co-delivering docetaxel and MMP-9 siRNA plasmid for nasopharyngeal carcinoma therapy in vivo. Mol Med Rep. 2017 Jun 07;: Authors: Liu T, Wu X, Wang Y, Hou X, Jiang G, Wu T, Xie H, Xie M Abstract The co-delivery of a drug and a target gene has become a primary strategy in cancer therapy. Based on our previous study, a synthesized star‑shaped co‑polymer consisting of β‑cyclodextrin (CD) and a poly(L‑lysine) dendron (PLLD) was used to co-deliver docetaxel (DOC) and matrix metalloproteinase 9 (MMP‑9) small interfering RNA, via CD‑PLLD/DOC/MMP‑9 complexes, into mice implanted with HN...

Targeting the Vav3 oncogene enhances docetaxel-induced apoptosis through the inhibition of androgen receptor phosphorylation in LNCaP prostate cancer cells under chronic hypoxia
Conclusions: Interrupting Vav3 signaling enhances docetaxel-induced apoptosis in LNCaP cells under chronic hypoxia by inhibiting the PI3K/Akt, ERK, and AR signaling pathways. Therapy targeting Vav3 in combination with docetaxel may have practical implications for managing castration-resistant prostate cancer.

Abstract 5403: Metronomic docetaxel in PRINT nanoparticles and EZH2 silencing have synergistic antitumor effect in ovarian cancer
Purpose: The purpose of this study was to investigate the combined antitumor effects of metronomic docetaxel-containing PLGA-PRINT nanoparticles and antiangiogenic mEZH2 siRNA incorporated into chitosan nanoparticles (CH-mEZH2 siRNA). Method: In vivo dose-finding studies and therapeutic experiments with PLGA-PRINT-docetaxel and CH-mEZH2 siRNA were conducted in well-established orthotopic mouse models of ovarian cancer (HeyA8 and SKOV3ip1). Antitumor effects were quantified through mean tumor weight and tumor nodule counts. Tumor tissues from these studies were stained to evaluate the proliferation index (Ki67), apoptosis i...

Bone-targeted nanoplatform enables efficient modulation of bone tumor microenvironment for prostate cancer bone metastasis treatment
Drug Deliv. 2022 Dec;29(1):889-905. doi: 10.1080/10717544.2022.2050845.ABSTRACTAs there is currently no effective therapy for patients with prostate cancer (PCa) bone metastasis, it was stringent to explore the relevant treatment strategies. Actually, the interaction between cancer cells and bone microenvironment plays important role in prostate cancer bone metastasis, especially the Sonic hedgehog protein (SHH) signaling in the bone microenvironment. The SHH promotes osteoblast maturation and osteoblast then secretes RANKL to induce osteoclastogenesis. Herein, this study develops bone-targeting calcium phosphate lipid hyb...
