STAT3 suppresses the AMPK α/ULK1-dependent induction of autophagy in glioblastoma cells
In this study, we found Serine-727 and Tyrosine-705 phosphorylation of STAT3 was constitutive in GBM cell lines. Tyrosine phosphorylation of STAT3 in GBM cells suppresses autophagy, whereas knockout (KO) of STAT3 increases ULK1 gene expression, increases TSC2-AMPKα-ULK1 signalling, and increases lysosomal Cathepsin D processing, leading to the stimulation of autophagy. Rescue of STAT3-KO cells by the enforced expression of wild-type (WT) STAT3 reverses these pathways and inhibits autophagy. Upregulation of atypical cadherin FAT1 promotes an immunosuppressive tumor microenvironment via TGF- β
Atypical cadherin FAT1 promotes glioblastoma (GBM) by promoting pro-tumorigenic inflammatory cytokine expression in tumor cells. However, tumors also have an immunosuppressive microenvironment, maintained by mediators like TGF-β cytokines. Here, we have studied the role of FAT1 in tumor immune suppression. Our preliminary TIMER2.0 analysis of TCGA database revealed inverse correlation of FAT1 expression with infiltration of tumor-inhibiting immune cells (like monocytes, T-cells, etc.) and positive correlation with tumor-promoting immune cells (like myeloid-derived suppressor cells/MDSCs) in various cancers. ...

The Neurokinin-1 Receptor Is Essential for the Viability of Human Glioma Cells: A Possible Target for Treating Glioblastoma
CONCLUSIONS: Our results show for the first time that the expression of the TAC1R gene (NK-1R) is essential for the viability of GAMG and U-87 MG glioma cells.
