Changes of flavin-containing monooxygenases and trimethylamine-N-oxide may be involved in the promotion of non-alcoholic fatty liver disease by intestinal microbiota metabolite trimethylamine
Conclusion, high fat and TMA could induce the expression of FMO1 and its metabolite TMAO. When FMO1 is silenced, the effects of high fat and TMA on TMAO are blocked. And the role of TMAO in NAFLD may be through the activation of UPR.PMID:35065293 | DOI:10.1016/j.bbrc.2022.01.060

Ursodesoxycholic acid is an FFA4 agonist and reduces hepatic steatosis via FFA4 signaling
This study not only identified a new skeleton of FFA4 agonists, but also demonstrated that FFA4 signal was accounting for the protective effects of UDCA in the NAFLD treatment.PMID:35033554 | DOI:10.1016/j.ejphar.2022.174760

TMEM88 Modulates Lipid Synthesis and Metabolism Cytokine by Regulating Wnt/ β-Catenin Signaling Pathway in Non-Alcoholic Fatty Liver Disease
Conclusion: Overall, the study proved that TMEM88 takes part in regulating the secretion of lipid synthesis and metabolism cytokine through the Wnt/β-catenin signaling pathway in AML-12 cells. Therefore, TMEM88 may be involved in the progress of NAFLD. Further research will bring new ideas for the study of NAFLD.

Vitexin attenuates autoimmune hepatitis in mouse induced by syngeneic liver cytosolic proteins via activation of AMPK/AKT/GSK-3 β/Nrf2 pathway
In conclusion, vitexin ameliorated hepatic injury in EAH mice through activation of the AMPK/AKT/GSK-3β pathway and upregulation of the Nrf2 gene.PMID:34953801 | DOI:10.1016/j.ejphar.2021.174720

Silibinin relieves UVB-induced apoptosis of human skin cells by inhibiting the YAP-p73 pathway
In this study we investigated the molecular mechanisms underlying. Human nonmalignant immortalized keratinocyte HaCaT cells and neonatal human foreskin fibroblasts HFFs were exposed to UVB irradiation. We showed that pre-treatment with silibinin dose-dependently decreased UVB-induced apoptosis of HaCaT cells. Furthermore, we showed that silibinin treatment inhibited nuclear translocation of YAP after UVB irradiation. Molecular docking analysis and DARTS assay confirmed the direct interaction of silibinin with YAP. Silencing YAP by siRNA had no influence on the survival of HaCaT cells, whereas inhibiting classical YAP-TEAD ...

Anti-inflammatory and anti-fibrotic effects of modafinil in nonalcoholic liver disease
Biomed Pharmacother. 2021 Dec;144:112372. doi: 10.1016/j.biopha.2021.112372. Epub 2021 Oct 28.ABSTRACTSmall- and intermediate-conductance Ca2+-activated K+ channels, KCa2.3 and KCa3.1, are involved in cellular signaling processes associated with inflammation and fibrosis. KCa2.3 and KCa3.1 are upregulated by proinflammatory cytokines and profibrotic growth factors. Cyclic AMP, which downregulates KCa2.3 and KCa3.1, is elevated by modafinil in cells; accordingly, we investigated whether modafinil exerts anti-inflammatory and anti-fibrotic responses via KCa2.3- and KCa3.1-mediated pathways in high-fat diet (HFD)- or thioacet...

Nimbolide attenuated the inflammation in the liver of autoimmune hepatitis's mice through regulation of HDAC3
Toxicol Appl Pharmacol. 2021 Nov 12:115795. doi: 10.1016/j.taap.2021.115795. Online ahead of print.ABSTRACTA chronic liver disease named autoimmune hepatitis (AIH) will carry elevated levels of inflammatory cytokines, but there is currently no effective treatment to cure it. Histone deacetylase 3 (HDAC3) takes an important position in regulating the expression of inflammatory genes. Nimbolide (NIB) is a limonoid extracted from the neem tree (Azadirachta indica) that has been found to be effective against many diseases, including cancer, scleroderma, and acute respiratory distress syndrome. Here, we investigated the protect...
