Blocking autophagy by the two-pore channels antagonist tetrandrine improves sorafenib-induced death of hepatocellular carcinoma cells
Toxicol In Vitro. 2023 Apr 28:105603. doi: 10.1016/j.tiv.2023.105603. Online ahead of print.ABSTRACTSorafenib, an oral multi-kinase inhibitor, used to treat hepatocellular carcinoma (HCC). However, drug resistance is still common in several HCC patients. This complex mechanism is not yet fully elucidated, driving the search for new therapeutic targets to potentiate the antitumoral effect of sorafenib. Recent findings have linked the expression of Two-Pore Channels (TPCs) receptors with the development and progression of cancer. TPCs receptors are stimulated by NAADP, a Ca2+ messenger, and inhibited by their antagonists Ned...

Epigenetic regulation of DHRS2 by SUV420H2 inhibits cell apoptosis in renal cell carcinoma
In this study, we found that SUV420H2 is overexpressed in renal cancers and that high SUV420H2 expression is associated with a poor prognosis, as evidenced by RCC RNA-seq results derived from the TCGA. SUV420H2 knockdown using siRNA led to growth suppression and cell apoptosis in the A498 cell line. Furthermore, we identified DHRS2 as a direct target of SUV420H2 in the apoptosis process through a ChIP assay with a histone 4 lysine 20 (H4K20) trimethylation antibody. Rescue experiments showed that cotreatment with siSUV420H2 and siDHRS2 attenuated cell growth suppression induced by SUV420H2 knockdown only. Additionally, tre...

Inhibition Mechanism of PinX1 Gene on Cancer Stem Cells of Nasopharyngeal Carcinoma
In this study, we found that the telomerase activity of the CD133 - + NC group was 1.001 ± 0.086, the CD133 - + pinx1sirna group was 0.974 ± 0.046, CD133+ + vector group was 0.928 ± 0.102, CD133+ + over PinX1 group was 0.703 ± 0.086. Therefore, the PinX1 gene can inhibit NPC stem cells by inhibiting telomerase activity.PMID:37114251 | DOI:10.14715/cmb/2022.68.10.28

Downregulation of ROR2 attenuates LPS ‐induced A549 cell injury through JNK and ERK signaling pathways
ConclusionsThus, the present data indicate that downregulation of ROR2 may decrease LPS-induced inflammatory responses and cell apoptosis through inhibiting JNK and ERK signaling pathway, which attenuates ALI.

Knockdown of LPCAT1 Repressed Hepatocellular Carcinoma Growth and Invasion by Targeting S100A11
CONCLUSION: LPCAT1 promoted the growth, invasion and migration of HCC cells by regulating S100A11 and Snail. Therefore, LPCAT1 may serve as a potential molecular target for the diagnosis and treatment of HCC.PMID:37094849
