Epigenetic regulation of epithelial-mesenchymal transition during cancer development
Int Rev Cell Mol Biol. 2023;380:1-61. doi: 10.1016/bs.ircmb.2023.05.007. Epub 2023 Jun 13.ABSTRACTEpithelial-mesenchymal transition (EMT) plays essential roles in promoting malignant transformation of epithelial cells, leading to cancer progression and metastasis. During EMT-induced cancer development, a wide variety of genes are dramatically modified, especially down-regulation of epithelial-related genes and up-regulation of mesenchymal-related genes. Expression of other EMT-related genes is also modified during the carcinogenic process. Especially, epigenetic modifications are observed in the EMT-related genes, indicati...

Acetyl-cinobufagin suppresses triple-negative breast cancer progression by inhibiting the STAT3 pathway
CONCLUSIONS: The findings of this study support the potential clinical use of acetyl-cinobufagin as a STAT3 inhibitor in TNBC adjuvant therapy.PMID:37651362 | DOI:10.18632/aging.204967

Impacts of LncRNA NORAD on the Proliferation, Apoptosis, and Chemosensitivity of Non-small Cell Lung Cancer Cells by Regulating ZNF217 through MiR-199a-3p
CONCLUSIONS: Down-regulating NORAD expression can enhance miR-199a-3p expression and inhibit ZNF217 expression, thereby inhibiting H460/DDP cell proliferation, promoting apoptosis and enhancing its DDP chemotherapy sensitivity.PMID:37653011 | DOI:10.3779/j.issn.1009-3419.2023.102.27

A combination of a cell penetrating peptide and a protein translation inhibitor kills metastatic breast cancer cells
Cell Death Discov. 2023 Aug 31;9(1):325. doi: 10.1038/s41420-023-01627-3.ABSTRACTCell Penetrating Peptides (CPPs) are promising anticancer and antimicrobial drugs. We recently reported that a peptide derived from the human mitochondrial/ER membrane-anchored NEET protein, Nutrient Autophagy Factor 1 (NAF-1; NAF-144-67), selectively permeates and kills human metastatic epithelial breast cancer cells (MDA-MB-231), but not control epithelial cells. As cancer cells alter their phenotype during growth and metastasis, we tested whether NAF-144-67 would also be efficient in killing other human epithelial breast cancer cells that m...

Evaluation of follow-up strategies for women with epithelial ovarian cancer following completion of primary treatment
CONCLUSIONS: Limited evidence from one trial suggests that routine surveillance with CA125 in asymptomatic women and treatment at CA125-defined relapse does not seem to offer survival advantage when compared to treatment at symptomatic relapse. However, this study pre-dates the use of PARPi maintenance treatment and the increased use of secondary cytoreductive surgery, so the results may be limited in their applicability to current practice. Limited evidence from one trial suggests that individualised nurse-led follow-up may improve HRQOL in women with ovarian cancer following completion of primary treatment. Large RCTs ar...

Wound-healing plasticity enables clonal expansion of founder progenitor cells in colitis
Dev Cell. 2023 Aug 25:S1534-5807(23)00407-0. doi: 10.1016/j.devcel.2023.08.011. Online ahead of print.ABSTRACTChronic colonic injury and inflammation pose high risks for field cancerization, wherein injury-associated mutations promote stem cell fitness and gradual clonal expansion. However, the long-term stability of some colitis-associated mutational fields could suggest alternate origins. Here, studies of acute murine colitis reveal a punctuated mechanism of massive, neutral clonal expansion during normal wound healing. Through three-dimensional (3D) imaging, quantitative fate mapping, and single-cell transcriptomics, we...

Flavonoids Quercetin and Kaempferol Are NR4A1 Antagonists and Suppress Endometriosis in Female Mice
Endocrinology. 2023 Aug 31:bqad133. doi: 10.1210/endocr/bqad133. Online ahead of print.ABSTRACTNuclear receptor 4A1 (NR4A1) plays an important role in endometriosis progression; levels of NR4A1 in endometriotic lesions are higher than in normal endometrium, and substituted bis-indole analogs (NR4A1) antagonists suppress endometriosis progression in mice with endometriosis. In addition, flavonoids kaempferol and quercetin are natural products that directly bind NR4A1 and significantly repress the intrinsic NR4A1-dependent transcriptional activity in human endometriotic epithelial and stromal cells and Ishikawa endometrial c...
