The Role of IgA in the Manifestation and Prevention of Allergic Immune Responses
AbstractPurpose of ReviewImmunoglobulin A (IgA) mediates immune exclusion of antigens in the gut. Notably, IgA plays also a role in the prevention of IgE-mediated allergies and induction of immune tolerance. The present review addresses the role of IgA in the manifestation of IgE-mediated allergies, including allergen-specific immunotherapy (AIT), the regulation of IgA production, and the mechanism of IgA in immune cell activation.Recent FindingsThe majority of studies report an association of IgA with the induction of immune tolerance in IgE-mediated allergies. However, reports on the involvement of humoral and mucosal Ig...

Modulation of immune response by nanoparticle-based immunotherapy against food allergens
The increasing prevalence of food allergies worldwide and the subsequent life-threatening anaphylactic reactions often have sparse treatment options, providing only symptomatic relief. Great strides have been made in research and in clinics in recent years to offer novel therapies for the treatment of allergic disorders. However, current allergen immunotherapy has its own shortcomings in terms of long-term efficacy and safety, due to the local side effects and the possibility of anaphylaxis. Allergen-specific immunotherapy is an established therapy in treating allergic asthma, allergic rhinitis, and allergic conjunctivitis...

An Overview of Nanotechnological Approaches for the Diagnosis and Treatment of Allergic Illness
Curr Pharm Des. 2023 Aug 28. doi: 10.2174/1381612829666230828104015. Online ahead of print.ABSTRACTAllergies are a major health issue. Allergen avoidance, antihistamines, and corticosteroids do not treat the pathology's causes, therefore long-term therapy is essential. Long-term allergen-specific immunotherapy builds immune tolerance to the allergen. Unfortunately, immunotherapies for all allergens are not available, and adverse reactions during therapy, especially in severely allergic persons, remain a worry. In this regard, cell and bio- or nanomaterial-based allergy treatments are promising. This overview covers the mos...

Anti-allergen monoclonal antibodies for the treatment of allergies
ConclusionThe development of anti-allergen antibody therapies offers great potential for the treatment of allergies. Of interest, the efficacy of passive immunization approaches will likely extend beyond allergen neutralization as in the presence of the allergen they might contribute to long-lasting disease modification.

Food sensitized pediatric patients show colonic cow's milk protein-specific Th2 cells
In conclusion, we isolated and characterized cow´s milk-specific lamina propria CD4+ Th2 cells from colonic inflammatory polyps. CCR9 expression on these cells, along with increase secretion of CCL25 in the polyp, favor recruitment and cow's milk specific allergic response within the inflammatory polyp tissue. Our findings may be critical to understand the underlying mechanism that promotes IgE synthesis in the colon of cow´s milk proteins allergic patients, contributing to the development of novel T cell-targeted immunotherapies.PMID:37478370 | DOI:10.1093/jleuko/qiad083

Update on oral and epicutaneous immunotherapy for children with food allergy
ConclusionThe implementation of recent treatment protocols has significantly improved the safety of OIT. Therefore, its continuation using safety protocols can be considered in the selection of the optimal OIT for individual patients, as provision of safe and effective treatment is crucial for the success of OIT.

Potential role for oral tolerance in gene therapy
Cell Immunol. 2023 Jun 28:104742. doi: 10.1016/j.cellimm.2023.104742. Online ahead of print.ABSTRACTOral immunotherapies are being developed for various autoimmune diseases and allergies to suppress immune responses in an antigen-specific manner. Previous studies have shown that anti-drug antibody (inhibitor) formation in protein replacement therapy for the inherited bleeding disorder hemophilia can be prevented by repeated oral delivery of coagulation factor antigens bioencapsulated in transplastomic lettuce cells. Here, we find that this approach substantially reduces antibody development against factor VIII in hemophili...
