Metformin ameliorates endometrial thickness in a rat model of thin endometrium
In conclusion, the study demonstrated that metformin ameliorates endometrial thickness in a rat model of thin endometrium by increasing endometrial proliferation and angiogenesis, without restoration of embryo implantation.PMID:38621769 | DOI:10.1111/1440-1681.13862 (Source: Clinical and Experimental Pharmacology and Physiology)

Weight Management with Orlistat in Type 2 Diabetes - An Electronic Healthcare Records Study
CONCLUSIONS: Orlistat was significantly associated with weight loss in T2DM/prediabetes when taken for at least 12 weeks. However, orlistat is infrequently prescribed and often taken for <12 weeks. Orlistat may be a useful adjunct to lifestyle modifications in T2DM/prediabetes however barriers to continuing orlistat means it may not be effective for everyone in managing weight loss.PMID:38621802 | DOI:10.3399/BJGP.2023.0684 (Source: The British Journal of General Practice)

Cardiovascular safety of evogliptin dual and triple therapy in patients with type 2 diabetes: a nationwide cohort study
Conclusions These findings suggest that dual or triple therapies of evogliptin for the management of type 2 diabetes in routine clinical practice present no cardiovascular harms, but could alternatively offer cardiovascular benefits in this patient population. (Source: BMJ Open)

Role of epigenetics in the regulation of skin aging and geroprotective intervention: A new sight
Biomed Pharmacother. 2024 Apr 13;174:116592. doi: 10.1016/j.biopha.2024.116592. Online ahead of print.ABSTRACTMultiple epigenetic factors play a regulatory role in maintaining the homeostasis of cutaneous components and are implicated in the aging process of the skin. They have been associated with the activation of the senescence program, which is the primary contributor to age-related decline in the skin. Senescent species drive a series of interconnected processes that impact the immediate surroundings, leading to structural changes, diminished functionality, and heightened vulnerability to infections. Geroprotective me...

< em > Ianus Bifrons < /em > : The Two Faces of Metformin
Cancers (Basel). 2024 Mar 26;16(7):1287. doi: 10.3390/cancers16071287.ABSTRACTThe ancient Roman god Ianus was a mysterious divinity with two opposite faces, one looking at the past and the other looking to the future. Likewise, metformin is an "old" drug, with one side looking at the metabolic role and the other looking at the anti-proliferative mechanism; therefore, it represents a typical and ideal bridge between diabetes and cancer. Metformin (1,1-dimethylbiguanidine hydrochloride) is a drug that has long been in use for the treatment of type 2 diabetes mellitus, but recently evidence is growing about its potential use ...

Synergistic effects of Metformin and Forskolin on oxidative stress induced by diabetes and hepatocellular cancer: an animal study
CONCLUSION: Metformin and Forskolin reduced oxidative stress in diabetic and HCC-induced rats. The results indicated that the combination of agents (Metformin & Forskolin) had greater therapeutic effects than Forskolin alone in reducing glucose levels in diabetic rats. However, the ameliorative effects of combining Metformin and Forskolin on blood and urine oxidative stress were not statistically higher than those of Metformin alone.PMID:38614244 | DOI:10.1016/j.toxicon.2024.107720 (Source: Toxicon)

Energy metabolism as therapeutic target for aged wound repair by engineered extracellular vesicle
Sci Adv. 2024 Apr 12;10(15):eadl0372. doi: 10.1126/sciadv.adl0372. Epub 2024 Apr 12.ABSTRACTAging skin, vulnerable to age-related defects, is poor in wound repair. Metabolic regulation in accumulated senescent cells (SnCs) with aging is essential for tissue homeostasis, and adequate ATP is important in cell activation for aged tissue repair. Strategies for ATP metabolism intervention hold prospects for therapeutic advances. Here, we found energy metabolic changes in aging skin from patients and mice. Our data show that metformin engineered EV (Met-EV) can enhance aged mouse skin repair, as well as ameliorate cellular senes...

Statin use as a moderator on the association between metformin and breast cancer risk in women with type 2 diabetes mellitus
ConclusionsExtending metformin or statin usage by one year conferred breast cancer protection in women with T2DM. Enhanced protective effect of metformin was observed among those who also use statins. These results suggest the potential of combined metformin and statin therapy as promising breast cancer prevention strategies. (Source: Cancer and Metabolism)

Safety vs price in the generic drug market: metformin
CONCLUSIONS: Recalls of metformin ER medications were effective in lowering NDMA levels below the FDA threshold; however, some samples of generic metformin still contained carcinogens even after FDA-announced recalls. The absence of any correlation with price indicates that potentially safer products are available on the market for the same price as poorer-quality products.PMID:38603530 | DOI:10.37765/ajmc.2024.89450 (Source: The American Journal of Managed Care)
