Mathematical models of coagulation - Are we there yet?
CONCLUSIONS: This systematic assessment of models of coagulation, using large dataset inputs, points to ways in which these models can be improved. A model that accurately reflects the effects of the multiple, subtle variations in an individual's haemostatic profile could be used for assessing antithrombotics or as a tool for precision medicine.PMID:38521192 | DOI:10.1016/j.jtha.2024.03.009 (Source: Thrombosis and Haemostasis)

Nonacog beta pegol prophylaxis in children with hemophilia B: safety, efficacy, and neurodevelopmental outcomes for up to 8 years
CONCLUSION: These data provide the longest follow-up for an extended half-life FIX and confirm the long-term efficacy of N9-GP prophylaxis in children with HB with no observed neurocognitive or neurologic safety concerns.PMID:38516633 | PMC:PMC10955654 | DOI:10.1016/j.rpth.2024.102341 (Source: Thrombosis and Haemostasis)

Standardisation of Definition and Management for Bleeding Disorder of Unknown Cause: Communication from the SSC of the ISTH
J Thromb Haemost. 2024 Mar 20:S1538-7836(24)00163-6. doi: 10.1016/j.jtha.2024.03.005. Online ahead of print.ABSTRACTIn many patients referred with significant bleeding phenotype, laboratory testing fails to define any hemostatic abnormalities. Clinical practise with respect to diagnosis and management of this patient cohort poses significant clinical challenges. We recommend that bleeding history in these patients should be objectively assessed using the ISTH bleeding assessment tool (BAT). Patients with increased BAT scores should progress to hemostasis laboratory testing. To diagnose BDUC, normal complete blood count, pr...

Recent Advances in Vascular Thiol Isomerases and Redox Systems in Platelet Function and Thrombosis
J Thromb Haemost. 2024 Mar 20:S1538-7836(24)00166-1. doi: 10.1016/j.jtha.2024.03.008. Online ahead of print.ABSTRACTThere have been substantial advances in vascular protein disulfide isomerases (PDIs) in platelet function and thrombosis in recent years. There are four known prothrombotic thiol isomerases, PDI, ERp57, ERp72, ERp46 and one antithrombotic PDI, transmembrane protein 1 (TMX1). A sixth PDI, ERp5, may exhibit either pro- or anti-thrombotic properties in platelets. Studies on ERp46 in platelet function and thrombosis provide insight into the mechanisms by which these enzymes function. ERp46-catalyzed disulfide cle...

Endothelial LAT1 (SLC7A5) Mediates S-Nitrosothiol Import and Modulates Respiratory Sequelae of Red Blood Cell Transfusion In Vivo
CONCLUSION: This is the first study showing a role for SNO transport by LAT1 in ECs in a genetic mouse model. We provide the first direct evidence for the coordination of RBC SNO export with EC SNO import via LAT1. SNO flux via LAT1 modulates RBC-EC sequestration in lungs after transfusion, and its disruption impairs blood oxygenation by the lung.PMID:38519039 | DOI:10.1055/s-0044-1782182 (Source: Thrombosis and Haemostasis)
