Human platelets contain a pool of free zinc in dense granules
CONCLUSION: We conclude that normal human platelets contain a pool of free Zn2+ concentrated in dense granules that is available for secretion upon platelet activation and potentially contributes to hemostasis.PMID:38481951 | PMC:PMC10933575 | DOI:10.1016/j.rpth.2024.102352 (Source: Thrombosis and Haemostasis)

Whole F8 gene sequencing identified pathogenic structural variant in the remaining unsolved patients with severe Haemophilia A
CONCLUSION: All the genetically unsolved severe haemophilia A of this cohort were due to structural variant disrupting the F8. This study highlighted the effectiveness of whole F8 sequencing to improve the molecular diagnosis of HA when the conventional approach has failed.PMID:38484912 | DOI:10.1016/j.jtha.2024.03.002 (Source: Thrombosis and Haemostasis)

Proportion of venous thromboembolism attributed to recognized prothrombotic genotypes in men and women
CONCLUSION: Our findings in a Norwegian population suggest that 52% and 38% of the VTEs can be attributed to known prothrombotic genotypes in men and women, respectively.PMID:38476459 | PMC:PMC10926210 | DOI:10.1016/j.rpth.2024.102343 (Source: Thrombosis and Haemostasis)

Performance Validation of the 2023 ACR/EULAR Antiphospholipid Syndrome Classification Criteria in an Antiphospholipid Syndrome Cohort
CONCLUSIONS: The 2023 criteria offer higher overall specificity and improved sensitivity in specific patient subsets, such as those with SLE, microvascular manifestations, cardiac valve disease, and thrombocytopenia when compared to the 2006 criteria.PMID:38462219 | DOI:10.1016/j.jtha.2024.02.019 (Source: Thrombosis and Haemostasis)

A novel method to quantify fibrin-fibrin and fibrin- α < sub > 2 < /sub > AP cross-links in thrombi formed from human trauma patient plasma
CONCLUSIONS: We have developed a novel method that allows us to quantify fibrin cross-links in trauma patients who have received TXA, providing an indirect measure of fibrinolytic resistance. Using this novel approach we have avoided the effect of TXA and shown that cryo increases fibrin-fibrin and fibrin-α2AP cross-linking when compared to Fg-C, highlighting the importance of FXIII in clot formation and stability in trauma patients.PMID:38462220 | DOI:10.1016/j.jtha.2024.03.001 (Source: Thrombosis and Haemostasis)

Emicizumab Promotes Factor Xa Generation on Endothelial Cells
CONCLUSIONS: Emicizumab promotes FXa generation on the surface of endothelial cells, which is markedly enhanced in the presence of FEIBA®. These findings demonstrate a potential mechanism for the thrombotic complications seen with the combined use of emicizumab and FEIBA®.PMID:38460838 | DOI:10.1016/j.jtha.2024.02.017 (Source: Thrombosis and Haemostasis)

Redox regulation of platelet function and thrombosis
J Thromb Haemost. 2024 Mar 7:S1538-7836(24)00122-3. doi: 10.1016/j.jtha.2024.02.018. Online ahead of print.ABSTRACTPlatelets are well-known players in several cardiovascular diseases such as atherosclerosis and venous thrombosis. There are increasing evidence demonstrating that reactive oxygen species (ROS) is generated within activated platelets. NADPH oxidase (NOX) is a major source of ROS generation in platelets. Ligand binding to platelet receptor glycoprotein VI (GPVI) stimulates intracellular ROS generation consisting of a Syk-independent production involving NOX activation and a following Syk-dependent generation. I...

Comprehensive Domain-Specific Analysis and IgG Profiling of anti-FVIII Antibodies using a bead-based multiplex immunoassay
CONCLUSION: LumiTope assay provides a sensitive and specific method for detection but also domain specification of anti-FVIII-antibodies. Implementation of bead-based assays could improve antibody detection, profiling, comparability of results, and can complement NBA.PMID:38453023 | DOI:10.1016/j.jtha.2024.02.016 (Source: Thrombosis and Haemostasis)

Mental health in persons with von Willebrand disease in the United States - A large national database study
CONCLUSIONS: Our study shows that mental health disorders in PwvWD are a significant health burden, and that burden is increased with documented bleeding symptoms. It is important that primary care physicians and hematologists caring for this population recognize this increased risk, and appropriately screen and refer to mental health professionals.PMID:38453024 | DOI:10.1016/j.jtha.2024.02.015 (Source: Thrombosis and Haemostasis)

Role of Microbiota-Derived Corisin in Coagulation Activation during SARS-CoV-2 Infection
CONCLUSION: The microbiota-derived corisin is significantly increased and correlated with activation of the coagulation system during SARS-CoV-2 infection, and corisin may directly increase the procoagulant activity in epithelial, endothelial and monocytic cells.PMID:38453025 | DOI:10.1016/j.jtha.2024.02.014 (Source: Thrombosis and Haemostasis)
