Association analysis of mitochondrial genome polymorphisms with backfat thickness in pigs
In this study, 243 pigs were collected to analyse the relationship between BFT and mtDNA polymorphisms. There were considerable differences in BFT, ranging from 5 mm to 18 mm. MtDNA D-loop sequencing discovered 48 polymorphic sites. Association analysis revealed that 30 variations were associated with BFT (P < 0.05). The polymorphism m.794A > G showed the maximum difference in BFT between A and G carriers, which differed at ∼2.5 mm (P < 0.001). The 48 polymorphic sites generated 22 haplotypes (H1-H22), which clustered into 4 haplogroups (HG1-HG4). HG1 had a lower BFT value than other three haplogroups (P < 0....

Comparative RNA isolation methods from fresh ejaculated spermatozoa in Sahiwal cattle ( < em > Bos indicus < /em > ) and Murrah buffalo ( < em > Bubalus bubalis < /em > ) bulls for high quality and enhanced RNA yield
Anim Biotechnol. 2023 Nov 15:1-12. doi: 10.1080/10495398.2023.2276713. Online ahead of print.ABSTRACTSperm mRNA transcriptional profiling can be used to evaluate the fertility of breeding bulls. The aim of the study was to compare the modified RNA isolation methods for higher RNA yield and quality from freshly ejaculated sperm of cattle and buffalo bulls. Ten fresh ejaculates from each Sahiwal (n = 10 bulls × 10 ejaculates) and Murrah bulls (n = 10 bulls x 10 ejaculates) were used for RNA isolation. From the recovered live sperm, total sperm RNA was isolated by conventional methods (TRIzol, Double TRIzol), membrane-based ...

Differences in composition and diversity of rumen fungi in buffalo fed different diets
The objective of this study was to investigate rumen fungal diversity and the presence of aerobic fungi in buffalo fed on different diets. Three adult buffaloes were used as experimental animals. Alfalfa hay, oat hay, whole corn silage, sugarcane shoot silage, fresh king grass, dried rice straw, and five kinds of mixed diets with concentrate to roughage ratios of 20:80, 35:65, 50:50, 65:35, and 80:20 were used as the experimental diets. The experimental animals were fed different diets for 22 days. Rumen fluid was collected from the rumen fistula for ITS (Internal Transcribed Spacer) sequencing 2 h after feeding on the mor...

Genetic diversity and phylogenetic relationship estimation of Shanxi indigenous goat breeds using microsatellite markers
The objective of this study was to assess the genetic diversity, phylogenetic relationship and population structure of five goat breeds in Shanxi, China. High genetic diversities were found in the five populations, among which, Licheng big green goat (LCBG) has the highest genetic diversity, while Jinlan cashmere goat (JLCG) population has the lowest genetic diversity. Bottleneck analysis showed the absence of recent genetic bottlenecks in the five goat populations. Genetic differentiation analysis shows that the closest genetic relationship between LCBG and LLBG (Lvliang black goat) was found, and the genetic distance bet...

Exome-wide comparative analyses revealed differentiating genomic regions for performance traits in Indian native buffaloes
This study is a maiden attempt to delineate the important variants and unique genes through exome sequencing for milk yield, milk composition, fertility, and adaptation traits in Indian local breeds of buffalo. In the present study, whole exome sequencing was performed on Chhattisgarhi (n = 3), Chilika (n = 4), Gojri (n = 3), and Murrah (n = 4) buffalo breeds and after stringent quality control, 4333, 6829, 4130, and 4854 InDels were revealed, respectively. Exome-wide FST along 100-kb sliding windows detected 27, 98, 38, and 35 outlier windows in Chhattisgarhi, Chilika, Gojri, and Murrah, respectively. The comparative exom...
