Comparative transcriptome profile analysis of granulosa cells from porcine ovarian follicles during early atresia
In this study, we isolated the granulosa cells from porcine ovarian follicles (3-5 mm diameter) divided into healthy follicles (HFs) and early atretic follicles (EAFs). We applied high-throughput RNA sequencing to identify and compare differentially expressed genes (DEGs) between HFs and EAFs. A total of 31,694 genes were detected, of which 21,806 were co-expressed in six samples, and 243 genes (p < 0.05; FDR < 0.05) were differentially expressed (DEGs), including 123 downregulated and 120 upregulated in EAFs. GO analysis highlighted hormone metabolism, plasma membrane localization, and transporter activity. The path...

Analysis of OCT4 and PGP9.5 gene expression in prenatal and postnatal buffalo ( < em > Bubalus bubalis < /em > ) testes
This study aimed to investigate and characterize the spermatogonial stem cells (SSCs) in buffaloes at different stages of development, including prenatal, neonatal, prepubertal, and adult testes. We sought a comprehensive understanding of these cells through a combination of histological, immunohistochemical, and ultrastructural analyses. Specifically, we examined changes in the expression of two potential SSC markers, OCT4 and PGP9.5, using immunohistochemistry. Additionally, we conducted a real-time quantitative polymerase chain reaction (RT-qPCR) to assess the relative gene expression of OCT4 and PGP9.5. The relative ex...

< em > Orychophragmus violaceus < /em > and/or chicory forage affects performance, egg quality, sensory evaluation and antioxidative properties in native laying hens
In conclusion, the present study suggests that CC had a better effect on the performance of the native laying hens than OV. The OV and CC affected egg quality, while YC was increased in CC and OVC groups. The OVC improved YC and serum antioxidative properties of native laying hens without affecting the performance.PMID:38006583 | DOI:10.1080/10495398.2023.2286610 (Source: Animal Biotechnology)

Whole genome resequencing revealed genomic variants and functional pathways related to adaptation in Indian yak populations
This study provides comprehensive information about genomic variants and their annotation in Indian yak populations, thus would serve as a data resource for researchers working on the yaks. Furthermore, it could be well exploited for better yak conservation strategies by estimating population genetics parameters viz., effective population size, inbreeding, and observed and expected heterozygosity.PMID:38006247 | DOI:10.1080/10495398.2023.2282723 (Source: Animal Biotechnology)

Condition optimization for electroporation transfection in horse skeletal muscle satellite cells
In this study, we investigated the effects of voltage, pulse duration, and DNA dosage on horse skeletal muscle satellite cells' electroporation transfection efficiency using the eukaryotic expression plasmid Td Tomato-C1 (5.5 kb) encoding the red fluorescent protein gene mainly based on fluorescence-positive cell rate and cell survival rate. By comparison of different voltages, pulse durations, and DNA doses, horse skeletal muscle satellite cells have nearly 80% transfection efficiency under the condition of voltage 120 V, DNA dosage 7 µg/ml, and pulse duration 30 ms. This optimized electroporation condition would facilit...

Effect of food supplementation on < em > in  vitro < /em > embryo production and growth performance in prepubertal Nelore heifers
Anim Biotechnol. 2023 Nov 17:1-10. doi: 10.1080/10495398.2023.2279612. Online ahead of print.ABSTRACTIn vitro embryos production from prepubertal heifers can help contribute to breeding programs; however, strategies are necessary to increase their embryo production. The aim of this study was to investigate the effects of two nutritional plans on oocyte recovery, embryo production and growth performance of prepubertal Nelore heifers. Thirty-four Nelore heifers with age of 6.5 months were divided into two feeding treatments (NP1 and NP2). The NP1 diets served as the control and NP2 diets were formulated to contain an average...

Comparative RNA isolation methods from fresh ejaculated spermatozoa in Sahiwal cattle ( < em > Bos indicus < /em > ) and Murrah buffalo ( < em > Bubalus bubalis < /em > ) bulls for high quality and enhanced RNA yield
Anim Biotechnol. 2023 Nov 15:1-12. doi: 10.1080/10495398.2023.2276713. Online ahead of print.ABSTRACTSperm mRNA transcriptional profiling can be used to evaluate the fertility of breeding bulls. The aim of the study was to compare the modified RNA isolation methods for higher RNA yield and quality from freshly ejaculated sperm of cattle and buffalo bulls. Ten fresh ejaculates from each Sahiwal (n = 10 bulls × 10 ejaculates) and Murrah bulls (n = 10 bulls x 10 ejaculates) were used for RNA isolation. From the recovered live sperm, total sperm RNA was isolated by conventional methods (TRIzol, Double TRIzol), membrane-based ...
