The MAGMA pipeline for comprehensive genomic analyses of clinical < i > Mycobacterium tuberculosis < /i > samples
ConclusionThe MAGMA pipeline could facilitate the integration of WGS into clinical care as it generates clinically relevant data on drug resistance and phylogeny in an automated, standardized, and reproducible manner. (Source: PLoS Computational Biology)

Guiding pragmatic treatment choices for rifampicin-resistant tuberculosis in the absence of second-line drug susceptibility testing
Extract With recent expansion in the use of bedaquiline and limited access to accurate drug susceptibility testing (DST), we read with concern of examples of the high prevalence of bedaquiline resistance detected in Mycobacterium tuberculosis isolates [1]. Choosing the most effective regimen for treatment of drug-resistant tuberculosis (TB) is crucially important [2] and requires accurate and timely DST. In the past decade, there has been wide roll-out of rapid molecular tests that can identify M. tuberculosis and mutations conferring rifampicin resistance (RR). However, for potent second-line drugs, such as fluoroquinolon...

Minimal effective dose of bedaquiline administered orally and activity of a long acting formulation of bedaquiline in the murine model of leprosy
ConclusionWe demonstrated that the MED of BDQ administered orally againstM.leprae was 3.3 mg/kg in mice and BDQ did not add significantly to the efficacy of CFZ at the doses tested. BDQ-LA IM was similar or less active than BDQ administered orally at equivalent dosing and frequency but should be tested at higher dosing in order to reach equivalent exposure in further experiments. Variations of the Mycobacterium abscessus F-ATP synthase subunit a-c interface alter binding and potency of the anti-TB drug bedaquiline
Biochem Biophys Res Commun. 2023 Nov 20;690:149249. doi: 10.1016/j.bbrc.2023.149249. Online ahead of print.ABSTRACTThe anti-tuberculosis therapeutic bedaquiline (BDQ) is used against Mycobacterium abscessus. In M. abscessus BDQ is only bacteriostatic and less potent compared to M. tuberculosis or M. smegmatis. Here we demonstrate its reduced ATP synthesis inhibition against M. abscessus inside-out vesicles, including the F1FO-ATP synthase. Molecular dynamics simulations and binding free energy calculations highlight the differences in drug-binding of the M. abscessus and M. smegmatis FO-domain at the lagging site, where th...
