Applications of tissue-specific and cancer-selective gene promoters for cancer diagnosis and therapy
Adv Cancer Res. 2023;160:253-315. doi: 10.1016/bs.acr.2023.03.005. Epub 2023 Apr 20.ABSTRACTCurrent treatment of solid tumors with standard of care chemotherapies, radiation therapy and/or immunotherapies are often limited by severe adverse toxic effects, resulting in a narrow therapeutic index. Cancer gene therapy represents a targeted approach that in principle could significantly reduce undesirable side effects in normal tissues while significantly inhibiting tumor growth and progression. To be effective, this strategy requires a clear understanding of the molecular biology of cancer development and evolution and develo...

Head and neck cancer treatment in the era of molecular medicine
Adv Cancer Res. 2023;160:205-252. doi: 10.1016/bs.acr.2023.03.004. Epub 2023 Apr 12.ABSTRACTHead and neck cancers are a heterogeneous group of highly aggressive tumors and collectively represent the sixth most common cancer worldwide. Most head and neck cancers are squamous cell carcinomas (HNSCCs). Current multimodal treatment concepts combine surgery, chemotherapy, irradiation, immunotherapy, and targeted therapeutics. Recent scientific advancements have enabled a more precise molecular characterization of HNSCC and revealed novel therapeutic targets and prognostic/predictive biomarkers. Notably, HNSCC is characterized b...

Integrated clustering signature of genomic heterogeneity, stemness and tumor microenvironment predicts glioma prognosis and immunotherapy response
CONCLUSIONS: This study identified risk hub genes and established a risk model by analyzing the genomic heterogeneity, stemness, and immune microenvironment of glioma. Our findings will facilitate the diagnosis and prediction of glioma prognosis and may lead to potential treatment strategies for glioma.PMID:37698534 | DOI:10.18632/aging.205018

Prognostic and immunological roles of IL18RAP in human cancers
In this study, based on bioinformatics analysis, we discovered that IL18RAP is related to the human tumor microenvironment and promotes various immune cells infiltration. Additionally, the multiple immunofluorescence staining revealed that with the increased expression of IL18RAP, the number of infiltrated M1 macrophages increased. This finding was confirmed by coculture migration analysis using three human cancer cell lines (MDA-MB-231, U251, and HepG2) with IL18RAP knockdown. We discovered a positive link between IL18RAP and the majority of immunostimulators, immunoinhibitors, major histocompatibility complex (MHC) molec...

Re-routing the GPS directing immunotherapy in endometrial cancer
Clin Cancer Res. 2023 Sep 12. doi: 10.1158/1078-0432.CCR-23-1953. Online ahead of print.ABSTRACTMMR status alone is insufficient to guide the use of PD-(L)1 monotherapy in endometrial cancer patients. Additional biomarkers, including TMB and CPS score may help to identify patients with pMMR tumors with a high probability of benefit from PD-(L)1 monotherapy, and those with dMMR tumors who might require combination strategies.PMID:37698948 | DOI:10.1158/1078-0432.CCR-23-1953
