The impact of the codelivery of drug-siRNA by trimethyl chitosan nanoparticles on the efficacy of chemotherapy for metastatic breast cancer cell line (MDA-MB-231).
Authors: Eivazy P, Atyabi F, Jadidi-Niaragh F, Aghebati Maleki L, Miahipour A, Abdolalizadeh J, Yousefi M Abstract High-mobility group protein two (HMGA2), a nonhistone nuclear-binding protein and its downregulators; vimentin, matrix metallopeptidase-9 (MMP-9), and E-cadherin are shown to contribute to tumor progression and metastasis. Thus, in this study, we checked simultaneous delivery of HMGA-2 siRNA and the anticancer drug doxorubicin to enhance the anticancer treatment effects. For this purpose, we used MTT assay and real-time polymerase chain reaction (RT-PCR). Our results showed that dual delivery of Dox an...

Fluorescent peptide biosensor for monitoring CDK4/cyclin D kinase activity in melanoma cell extracts, mouse xenografts and skin biopsies
Publication date: 15 November 2016 Source:Biosensors and Bioelectronics, Volume 85 Author(s): Camille Prével, Morgan Pellerano, Juan A. González-Vera, Pauline Henri, Laurent Meunier, Julien Vollaire, Véronique Josserand, May C. Morris Melanoma constitutes the most aggressive form of skin cancer, which further metastasizes into a deadly form of cancer. The p16INK4a-Cyclin D-CDK4/6-pRb pathway is dysregulated in 90% of melanomas. CDK4/Cyclin D kinase hyperactivation, associated with mutation of CDK4, amplification of Cyclin D or loss of p16INK4a leads to increased risk of developing melanoma. This kinase therefo...

Lipid-based siRNA Delivery Systems: Challenges, Promises and Solutions Along the Long Journey.
Abstract RNA interference (RNAi) is an evolutionary conserved highly specific gene-silencing mechanism initiated by small interfering RNA (siRNA) molecules. Fast-paced preclinical and clinical studies helped the siRNA technology become an efficient tool for undruggable targets in different diseases including genetic diseases, viral diseases and cancer. Despite great feature of siRNAs that can down-regulate any protein in the cells, the full potential and the success of the preclinical studies could not be translated into largely successful clinical outcomes. It has become clear that the possibility of overcoming t...

Polyelectrolyte complexes of hTERT siRNA and polyethyleneimine: Effect of degree of PEG grafting on biological and cellular activity.
Authors: Safari F, Tamaddon AM, Zarghami N, Abolmali S, Akbarzadeh A Abstract Gene silencing by siRNA (short interfering RNA)-targeted human telomerase reverse transcriptase (hTERT) is considered a successful strategy for cancer gene therapy. Polyelectrolyte complexes (PEC) of siRNA and cationic polymers such as polyethyleneimine (PEI) have been widely used for cellular transfection; however, they demonstrate some disadvantages such as cytotoxicity and extracellular matrix restrictions. PEG grafting technology was used in an attempt to improve the biocompatibility of PECs. Considering that this technology may compr...

Preparation and characterization of silk fibroin/oligochitosan nanoparticles for siRNA delivery.
In this study, oligochitosan (OC) combined with silk fibroin (SF) was formulated and proposed as a novel carrier for siRNA. The obtained SF/OC/siRNA nanoparticles (NPs) were characterized according to their physicochemical properties, such as their size, zeta potential, loading efficiency, stability, cytotoxicity, cellular uptake and transfection efficiency, and their properties were compared with those of OC polyplexes. The mean diameter of SF/OC/siRNA NPs was not significantly different compared to polyplexes, and the particle size ranged between 250 and 450nm. Increased amounts of SF in NPs enhanced their loading effici...

Evaluation of the physicochemical properties and the biocompatibility of polyethylene glycol-conjugated gold nanoparticles: A formulation strategy for siRNA delivery.
Abstract The potential of RNA interference (RNAi)-based therapeutics for cancer has received much attention; however, delivery of RNAi effectors, such as small interfering RNA (siRNA), remains an obstacle to clinical translation. Non-viral delivery vectors have been used extensively to enhance siRNA delivery. Recently, the potential of gold nanoparticles (AuNPs) for transporting drugs, proteins and genetic materials has been demonstrated. Previously, our laboratory synthesised positively charged, surfactant-free AuNPs in water by the reduction of gold (III) chloride (AuCl3) using hydroxylamine hydrochloride (NH2OH...

Functional Assays for Specific Targeting and Delivery of RNA Nanoparticles to Brain Tumor
Cumulative progress in nanoparticle development has opened a new era of targeted delivery of therapeutics to cancer cells and tissue. However, developing proper detection methods has lagged behind resulting in the lack of precise evaluation and monitoring of the systemically administered nanoparticles. RNA nanoparticles derived from the bacteriophage phi29 DNA packaging motor pRNA have emerged as a new generation of drugs for cancer therapy. Multifunctional RNA nanoparticles can be fabricated by bottom-up self-assembly of engineered RNA fragments harboring targeting (RNA aptamer or chemical ligand), therapeutic (siRNA, miR...

Achieving successful delivery of oligonucleotides - from physico-chemical characterization to in vivo evaluation.
Abstract RNA interference is one of the most promising fields in modern medicine to treat several diseases, ranging from cancer to cardiac diseases, passing through viral infections and metabolic pathologies. Since the discovery of the potential therapeutic properties of non-self oligonucleotides, it was clear that it is important to develop delivery systems that are able to increase plasma stability and bestow membrane-crossing abilities to the oligonucleotides in order to reach their cytoplasmic targets. Polymer therapeutics, among other systems, are widely investigated as delivery systems for therapeutic agents...
