The potential role of mechanosensitive ion channels in substrate stiffness-regulated Ca < sup > 2+ < /sup > response in chondrocytes
CONCLUSIONS: Collectively, these findings indicate that chondrocytes might perceive and distinguish the different PCM stiffness by using different mechanosensitive ion channels.PMID:34814790 | DOI:10.1080/03008207.2021.2007902

Physalin A Inhibits MAPK and NF- κB Signal Transduction Through Integrin αVβ3 and Exerts Chondroprotective Effect
In conclusion, our work demonstrates that PA exhibits a chondroprotective effect that may be mediated by integrin αVβ3. Thus, PA or integrin αVβ3 might be a promising agent or molecular target for the treatment of OA.

Epigenetic Regulation of Nutrient Transporters in Rheumatoid Arthritis Fibroblast ‐Like Synoviocytes
ConclusionThis study demonstrates that changes in the epigenetic landscape of genes related to nutrient transporters and metabolic pathways identify RA-specific targets, including critical SLC transporters, enzymes, and transcription factors, to develop novel therapeutic agents. This article is protected by copyright. Pin1-mediated regulation of articular cartilage stem/progenitor cell aging
In this study, we investigated the role of Pin1 in the aging of rat knee joint CSPCs. We isolated CSPCs from rat knee joints and demonstrated that, in long-term in vitro culture, Pin1 protein levels are significantly reduced. At the same time, expression of the senescence-related β-galactosidase and the senescence marker p16INK4A were markedly elevated. In addition, Pin1 overexpression reversed the progression of cellular senescence, as evidenced by the down-regulation of senescence-related β-galactosidase, increased EdU positive cells and diminished levels of p16INK4A. In contrast, Pin1 siRNA incorporation promoted CSPC...

RPL38 knockdown inhibits the inflammation and apoptosis in chondrocytes through regulating METTL3-mediated SOCS2 m6A modification in osteoarthritis
ConclusionRPL38 knockdown inhibited osteoarthritic chondrocyte dysfunction and alleviated OA progression through promoting METTL3-m6A-mediated SOCS2 expression.

BNIP3 mediates the different adaptive responses of fibroblast-like synovial cells to hypoxia in patients with osteoarthritis and rheumatoid arthritis
CONCLUSION: In summary, RA-FLSs maintained intracellular redox balance through mitophagy to promote cell survival under hypoxia. The mitophagy of OA-FLSs was too little to maintain the redox balance of mitochondria, resulting in apoptosis. The difference of mitophagy between OA-FLSs and RA-FLSs under hypoxia is mediated by the level of BNIP3 expression.PMID:35690741 | DOI:10.1186/s10020-022-00490-9
