The Expression and Role of NADPH Oxidase 2 in Colon Cancer
CONCLUSION: NOX2 expression level has a close association with the prognosis of colon cancer patients and physiological functions of colon cancer cells. NOX2 may be a useful prognostic biomarker for colon cancer patients.PMID:37247898 | DOI:10.21873/anticanres.16427

Inhibition of ubiquitin specific peptidase 8 is effective against 5-fluorouracil resistance in colon cancer via suppressing EGFR and EGFR-mediated signaling pathways
CONCLUSIONS: Our work is the first to reveal the essential role of USP8 in colon cancer via EGFR oncogenic signalling pathways. Our findings provide a proof-of-concept that USP8 inhibitors are promising candidates to overcome 5-FU resistance in colon cancer.PMID:37222451 | DOI:10.14670/HH-18-629

Suppression of small nucleolar RNA host gene 8 (SNHG8) inhibits the progression of colorectal cancer cells
In this study, we explored the role of SNHG8 in CRC cell lines by performing a series of functional experiments. Similar to the data reported in the Encyclopedia of RNA Interactome, our RT-qPCR results showed that SNHG8 expression was significantly upregulated in CRC cell lines (DLD-1, HT-29, HCT-116, and SW480) compared to the normal colon cell line (CCD-112CoN). We performed dicer-substrate siRNA transfection to knockdown the expression of SNHG8 in HCT-116 and SW480 cell lines which were expressing high levels of SNHG8. SNHG8 knockdown significantly reduced CRC cell growth and proliferation by inducing autophagy and apop...

β-Sitosterol blocks the LEF-1-mediated Wnt/β-catenin pathway to inhibit proliferation of human colon cancer cells
CONCLUSION: According to our results, LEF-1 down-regulation can effectively block Wnt/β-catenin pathway, inhibit CC cell growth and migration. Collectively, β-sitosterol can be used to treat CC, which can provide anti-tumor activity by targeting LEF-1.PMID:36603684 | DOI:10.1016/j.cellsig.2022.110585

Hexavalent chromium induces centrosome amplification through ROS-ATF6-PLK4 pathway in colon cancer cells
In conclusion, our results suggest that hexavalent chromium induces CA via the ROS-ATF6-PLK4 pathway and provides molecular targets for inhibiting chromium-mediated CA, which may be useful for the assessment of CA in chromium-promoted tumorigenesis and cancer cell metastasis. Constitutive β-catenin overexpression represses lncRNA MIR100HG transcription via HDAC6-mediated histone modification in colorectal cancer
Mol Cancer Res. 2022 Mar 4:molcanres.0923.2021. doi: 10.1158/1541-7786.MCR-21-0923. Online ahead of print.ABSTRACTWnt/β-catenin signaling plays a critical role in colonic carcinogenesis. However, non-coding RNAs (ncRNAs) transcriptionally regulated by β-catenin are largely unknown. Herein, we found that lncRNA MIR100HG (lnc-MIR100HG) negatively correlated with target genes of β-catenin from The Cancer Genome Atlas colorectal carcinoma (CRC) database, which was verified in 48 paired CRC specimens. In addition, constitutive overexpression of β-catenin decreased primary and mature lnc-MIR100HG levels, while blockage of β...

LncRNA CCAT1 sponges miR-218-5p to promote EMT, cellular migration and invasion of retinoblastoma by targeting MTF2
In conclusion, here we identified the lncRNA CCAT1/miR-218-5p/MTF2 axis in RB cell lines. Our investigations on the function of lncRNA CCAT1 hint a novel biomarker and therapeutic option to facilitate RB diagnosis and treatment.PMID:34265414 | DOI:10.1016/j.cellsig.2021.110088

Responses of porcupine and Wntless proteins to oxidative, hypoxic and endoplasmic reticulum stresses
Cell Signal. 2021 May 17:110047. doi: 10.1016/j.cellsig.2021.110047. Online ahead of print.ABSTRACTThe WNT (Wingless and Int-1) proteins play a role in stem cell development and cell differentiation. Mutations in the WNT proteins lead to the development of various tumours, including gastric tumours. Porcupine (PORCN) is a palmitoyltransferase and Wntless (WLS) is a chaperone protein that modify and fold the WNT proteins respectively and are involved in their proper secretion and binding to the frizzled (FZD) receptor and the lipoprotein receptor-related protein 5 or 6 (LRP5/6). We investigated how modifications of PORCN an...
