PACSIN2 regulates platelet integrin β1 hemostatic function
CONCLUSIONS: Pacsin2-/- mice displayed severe thrombus formation defects due to hyperactive platelet integrin β1. The data suggest that PACSIN2 binding to FlnA negatively regulates platelet integrin β1 hemostatic function.PMID:37678551 | DOI:10.1016/j.jtha.2023.08.026

Post-mortem diagnosis of severe factor X deficiency in a fetus with intracranial haemorrhage resulting in intrauterine death
We describe a case of fetal death at 26 weeks due to massive ICH. Gene panel analysis of post-mortem samples revealed homozygosity for a pathological F10 gene variant (c.1210T>C, p.Cys404Arg), which impedes correct folding of the catalytic serine protease domain and therefore causes a significant reduction in FX levels. The parents, not consanguineous but of the same ethnic community, were found to be heterozygous for this variant and did not have any personal or family history of abnormal bleeding. This is, to the best of our knowledge, the first reported case of severe FX deficiency resulting in ICH diagnosed through ...

Platelets and neutrophils cooperate to induce increased NETosis in JAK2V617F myeloproliferative neoplasms
CONCLUSIONS: Our study demonstrates that JAK2V617F neutrophils alone are not sufficient to promote thrombosis; rather, platelets cooperate with neutrophils to promote NETosis in vivo. A new role for aspirin in thrombosis prevention in MPNs was also identified.PMID:37678548 | DOI:10.1016/j.jtha.2023.08.028

Mice possess a more limited natural anti-human Factor VIII antibody repertoire than humans that is produced disproportionately by marginal zone B cells
CONCLUSIONS: There is a preponderance of natural anti-hFVIII antibodies in both mice and healthy adult human donors.PMID:37678547 | DOI:10.1016/j.jtha.2023.08.033

Congenital thrombophilia in East-Asian venous thromboembolism population: a systematic review and meta-analysis
CONCLUSION: The prevalence of PC, PS, and AT deficiencies was relatively high, while a much lower prevalence of FVL and prothrombin G20210A mutations were identified in East-Asian patients with VTE. Our data stress the relative higher prevalence of PC, PS, and AT deficiencies for thrombophilia in the East-Asian VTE population.PMID:37674867 | PMC:PMC10477689 | DOI:10.1016/j.rpth.2023.102157

PAR4 Inhibition Reduces Coronary Artery Atherosclerosis and Myocardial Fibrosis in SR-B1/LDLR Double Knockout Mice
CONCLUSIONS: The PAR4 inhibitory RAG8 pepducin reduced coronary artery atherosclerosis and myocardial fibrosis in SR-B1/LDLR double knockout mice fed a high-fat, high-cholesterol diet containing cholate. Furthermore, RAG8 reduced VCAM-1 in nonatherosclerotic coronary arteries and reduced leukocyte and platelet accumulation in atherosclerotic coronary arteries. These findings identify PAR4 as an attractive target in reducing coronary artery disease development, and the use of RAG8 may potentially be beneficial in cardiovascular disease.PMID:37675637 | DOI:10.1161/ATVBAHA.123.319767

A bio-inspired nanoparticle coating for vascular healing and immunomodulatory by cGMP-PKG and NF-kappa B signaling pathways
Biomaterials. 2023 Aug 27;302:122288. doi: 10.1016/j.biomaterials.2023.122288. Online ahead of print.ABSTRACTDrug-eluting stents (DESs) implantation is an effective method to tackle in-stent restenosis (ISR), which has been considered as an efficient treatment for coronary atherosclerosis. Although fruitful results have been achieved in treating coronary artery diseases (CAD), concern has arisen regarding the long-term safety and efficacy of DESs, primarily due to adverse events such as delayed re-endothelialization, persistent inflammatory response, and late stent thrombosis (LST). Taking inspiration from the immunomodula...

Gas station in blood vessels: An endothelium mimicking, self-sustainable nitric oxide fueling stent coating for prevention of thrombosis and restenosis
Biomaterials. 2023 Aug 31;302:122311. doi: 10.1016/j.biomaterials.2023.122311. Online ahead of print.ABSTRACTStenting is the primary treatment for vascular obstruction-related cardiovascular diseases, but it inevitably causes endothelial injury which may lead to severe thrombosis and restenosis. Maintaining nitric oxide (NO, a vasoactive mediator) production and grafting endothelial glycocalyx such as heparin (Hep) onto the surface of cardiovascular stents could effectively reconstruct the damaged endothelium. However, insufficient endogenous NO donors may impede NO catalytic generation and fail to sustain cardiovascular h...

Characteristics and risk factors for advanced lung cancer with pulmonary embolism: A cross-sectional, case-control study
CONCLUSION: Our study provides valuable insights into the epidemiology and prognosis of PE in lung cancer patients and suggests that a poor ECOG PS, which has not been previously reported, is an independent risk factor for PE.PMID:37675831 | DOI:10.1111/crj.13692

Management of Paget-Schroetter Syndrome: a Systematic Review and Meta-Analysis
CONCLUSION: Non-operative management of PSS with AC alone results in persistent symptoms in 46% of patients, while 96% of patients managed with CDT + FFR are symptom free at end of follow up. Superiority of CDT + FRR compared with AC, CDT + AC, and AC + FRR was confirmed by meta-analysis. The overall quality of included reports was moderate, and the level of certainty was "very low".PMID:37678659 | DOI:10.1016/j.ejvs.2023.08.065

3D Printing-Assisted versus Conventional Extracorporeal Fenestration Tevar for Stanford Type B Arteries Dissection with Undesirable Proximal Anchoring Zone: Efficacy Analysis
CONCLUSIONS: Both techniques are safe and effective for TBAD with an undesirable proximal landing zone. The 3D-printing-assisted extracorporeal fenestration TEVAR technique has advantages in operative time, endoleak risk, and aortic remodeling, while the traditional extracorporeal fenestration TEVAR technique has advantages in stent modification.PMID:37679083 | DOI:10.59958/hsf.5885
