Phosphorylation of KRT8 (keratin 8) by excessive mechanical load-activated PKN (protein kinase N) impairs autophagosome initiation and contributes to disc degeneration
This study validates a protective role of Krt8 during overloading-induced IDD and demonstrates that targeting overloading activation of PKNs could be a novel and effective approach to mechano stress-induced pathologies with a wider window of therapeutic opportunity.Abbreviations: AAV: adeno-associated virus; AF: anulus fibrosus; ANOVA: analysis of variance; ATG: autophagy related; BSA: bovine serum albumin; cDNA: complementary deoxyribonucleic acid; CEP: cartilaginous endplates; CHX: cycloheximide; cKO: conditional knockout; Cor: coronal plane; CT: computed tomography; Cy: coccygeal vertebra; D: aspartic acid; DEG: differe...

SGLT2 inhibitor canagliflozin alleviates vascular calcification through suppression of NLRP3 inflammasome
CONCLUSIONS: Our study for the first time demonstrates that CANA exerts a protective effect on vascular calcification at least partially via suppressing NLRP3 signaling pathway. Therefore, supplementation of CANA as well as inhibition of NLRP3 inflammasome presents a potential therapy for vascular calcification.PMID:37523743 | DOI:10.1093/cvr/cvad119

Hyperglycaemia aggravates periodontal inflamm-aging by promoting SETDB1-mediated LINE-1 de-repression in macrophages
CONCLUSIONS: The findings of this study show that SETDB1 regulates senescence-like phenotypical switching of macrophages and is a potential candidate for the treatment of diabetes-induced periodontal inflamm-aging.PMID:37661095 | DOI:10.1111/jcpe.13871

Single photon emission computed tomography for the diagnosis of coronary artery disease: an evidence-based analysis.
The objective of the analysis is to determine the diagnostic accuracy of single photon emission tomography (SPECT) in the diagnosis of coronary artery disease (CAD) compared to the reference standard of coronary angiography (CA). The analysis is primarily meant to allow for indirect comparisons between non-invasive strategies for the diagnosis of CAD, using CA as a reference standard. SPECT: Cardiac SPECT, or myocardial perfusion scintigraphy (MPS), is a widely used nuclear, non-invasive image acquisition technique for investigating ischemic heart disease. SPECT is currently appropriate for all aspects of detecting and man...

Validity of neck circumference as a marker of adiposity in children and adolescents, and in adults: a systematic review.
In conclusion, neck circumference is associated with doubly-indirect markers of total and central fat mass in children and adolescents, while in adults it is associated with adiposity parameters measured with indirect and doubly-indirect methods. Further studies including methods that address the association between neck circumference and body composition analyzed using reference methods are required, especially in children and adolescents. 3D printing and its applications in orthopaedic trauma: A technological marvel
ConclusionAll over the world, orthopaedic Surgeon's and allied professionals and scientists are enthusiastically using 3D printing technology for designing patient specific models, instrumentation, implants, orthosis and prosthesis, besides 3D bioprinting of bone and cartilage scaffolding, and the same has been applied for nearly all areas of orthopaedic trauma surgery, from head to foot.

Bronchoscopy-guided antimicrobial therapy for cystic fibrosis.
CONCLUSIONS: This review, limited to a single, well-designed randomized controlled study, shows no clear evidence to support the routine use of bronchoalveolar lavage for the diagnosis and management of pulmonary infection in pre-school children with cystic fibrosis compared to the standard practice of providing treatment based on results of oropharyngeal culture and clinical symptoms. No evidence was available for adult and adolescent populations. PMID: 30221745 [PubMed - as supplied by publisher]

Treatment-Resistant Schizophrenia: Genetic and Neuroimaging Correlates
Conclusion and Future Perspectives Although interesting data have come from pharmacogenetics, neuroimaging and the interaction of both fields of study, few converging findings are available that describe the antipsychotic treatment response and resistance mechanisms in schizophrenia (DeLisi and Fleischhacker, 2016). Based on the available evidence, the results from both neuroimaging and pharmacogenetic/pharmacogenomic studies point to an overlap in the neurobiological vulnerability risk factors influencing the antipsychotic drug response in schizophrenia and the risk factors underlying schizophrenia itself. Currently for ...

Systematic Review and Meta-Analysis Confirms Significant Contribution of Surfactant Protein D in Chronic Obstructive Pulmonary Disease
Conclusion: Serum concentration and alleleic conformation of SFTPD has a significantly high predictive value for COPD and AECOPD. Thus, these can be tested further and could be applied as a predictive or prognostic marker. Introduction Chronic Obstructive Pulmonary Disease (COPD) affects lungs and exhibits irreversible airflow conditions that leads to improper respiratory function (Carolan et al., 2014). COPD is a global disease burden which accounts for ~3 million deaths annually (Zemans et al., 2017) and is responsible for the increase in worldwide mortality and morbidity (Dickens et al., 2011). Chronic O...

Fight Aging! Newsletter, May 4th 2020
The objective is to start treating chronic diseases from the root and not the symptoms of the disease. As we are starting to enroll patients in "senolytics-clinical trials," it will be imperative to assess if senolysis efficiently targets the primary cause of disease or if it works best in combination with other drugs. Additional basic science research is required to address the fundamental role of senescent cells, especially in the established contexts of disease. Chloroquine or hydroxychloroquine for prevention and treatment of COVID-19
CONCLUSIONS: HCQ for people infected with COVID-19 has little or no effect on the risk of death and probably no effect on progression to mechanical ventilation. Adverse events are tripled compared to placebo, but very few serious adverse events were found. No further trials of hydroxychloroquine or chloroquine for treatment should be carried out. These results make it less likely that the drug is effective in protecting people from infection, although this is not excluded entirely. It is probably sensible to complete trials examining prevention of infection, and ensure these are carried out to a high standard to provide un...
