A Pharmacovigilance Study of adverse events associated with polymyxins based on the U.S. Food and Drug Administration adverse event reporting system (FAERS) database
CONCLUSIONS: Patients treated with colistin displayed a higher potential risk of nephrotoxicity but a lower risk of neurotoxicity when compared to those treated with PMB. Clinicians should be vigilant in monitoring the AEs of hyperpigmentation disorders induced by PMB, as there have been reported cases associated with its use, whereas no instances of this AE were reported among colistin-treated patients. Nevertheless, further research, especially with larger sample sizes, is imperative to validate these findings.PMID:38676603 | DOI:10.1080/14740338.2024.2348610 (Source: Expert Opinion on Drug Safety)

GSE264574 Understanding resistance to polymyxin and amikacin in Klebsiella pneumoniae
Contributors : Camila de Souza ; Octavio FrancoSeries Type : Expression profiling by high throughput sequencingOrganism : Klebsiella pneumoniaeKlebsiella pneumoniae poses a significant global health threat primarily attributable to its pronounced resistance. Here, we report an in vitro acquired resistance analyses of K. pneumoniae to the combination of amikacin and polymyxin B. We found some differentially expressed genes associated with the resistome of K. pneumoniae. The main differences were found in the genes aphA, asmA, phoP, and in the arn operon. Once these genes are related to modification in lipopolysaccharides,...

The NLRP3 Inflammasome Is a Major Cause of Acute Renal Failure Induced by Polypeptide Antibiotics
J Immunol. 2024 Apr 19:ji2300193. doi: 10.4049/jimmunol.2300193. Online ahead of print.ABSTRACTDrug-induced acute renal failure (ARF) is a public health concern that hinders optimal drug therapy. However, pathological mechanisms of drug-induced ARF remain to be elucidated. Here, we show that a pathological process of drug-induced ARF is mediated by proinflammatory cross-talk between kidney tubular cells and macrophages. Both polymyxin B and colistin, polypeptide antibiotics, frequently cause ARF, stimulated the ERK and NF-κB pathways in kidney tubular cells, and thereby upregulated M-CSF and MCP-1, leading to infiltration...

Paper-based fluorescence sensor array with functionalized carbon quantum dots for bacterial discrimination using a machine learning algorithm
Anal Bioanal Chem. 2024 Apr 17. doi: 10.1007/s00216-024-05262-4. Online ahead of print.ABSTRACTThe rapid discrimination of bacteria is currently an emerging trend in the fields of food safety, medical detection, and environmental observation. Traditional methods often require lengthy culturing processes, specialized analytical equipment, and bacterial recognition receptors. In response to this need, we have developed a paper-based fluorescence sensor array platform for identifying different bacteria. The sensor array is based on three unique carbon quantum dots (CQDs) as sensing units, each modified with a different antibi...

A systematic evaluation of population pharmacokinetic models for polymyxin B in patients with liver and/or kidney dysfunction
This study aimed to systemically evaluate eleven PMB PopPK models from ten published literature based on a new independent population, which was divided into four different populations, patients with liver dysfunction, kidney dysfunction, liver and kidney dysfunction, and normal liver and kidney function. The whole data set consisted of 146 patients with 391 PMB concentrations. The prediction- and simulation-based diagnostics and Bayesian forecasting were conducted to evaluate model predictability. In the overall evaluation process, none of the models exhibited satisfactory predictive ability in both prediction- and simula...

Discovery of a novel class of rosmarinic acid derivatives as antibacterial agents: Synthesis, structure-activity relationship and mechanism of action
Bioorg Chem. 2024 Mar 28;146:107318. doi: 10.1016/j.bioorg.2024.107318. Online ahead of print.ABSTRACTTwenty-seven rosmarinic acid derivatives were synthesized, among which compound RA-N8 exhibited the most potent antibacterial ability. The minimum inhibition concentration of RA-N8 against both S. aureus (ATCC 29213) and MRSA (ATCC BAA41 and ATCC 43300) was found to be 6 μg/mL, and RA-N8 killed E. coli (ATCC 25922) at 3 μg/mL in the presence of polymyxin B nonapeptide (PMBN) which increased the permeability of E. coli. RA-N8 exhibited a weak hemolytic effect at the minimum inhibitory concentration. SYTOX Green assay, SEM...
