Acetaminophen overdose causes a breach of the blood-bile barrier in mice but not in rats
In conclusion, different mechanisms cause cell death in rats and mice, whereby oxidative stress and a breach of the blood-bile barrier are seen only in mice. Since transient cholestasis also occurs in human patients with APAP overdose, mice are a clinically relevant species to study APAP hepatotoxicity but not rats.PMID:38466352 | DOI:10.1007/s00204-024-03705-6 (Source: Archives of Toxicology)

Medication-overuse headache: A pharmacovigilance study in France
CONCLUSIONS: Certain drug classes show a high reporting rate of MOH and caution should be exercised when prescribing these drugs. Notably, warnings about MOH must be mentioned in the SmPC of all concerned drug classes.PMID:38458945 | DOI:10.1016/j.therap.2024.02.001 (Source: Therapie)

Preclinical efficacy and safety of encapsulated proliferating human hepatocyte organoids in treating liver failure
Cell Stem Cell. 2024 Feb 27:S1934-5909(24)00048-1. doi: 10.1016/j.stem.2024.02.005. Online ahead of print.ABSTRACTAlginate-encapsulated hepatocyte transplantation is a promising strategy to treat liver failure. However, its clinical application was impeded by the lack of primary human hepatocytes and difficulty in controlling their quality. We previously reported proliferating human hepatocytes (ProliHHs). Here, quality-controlled ProliHHs were produced in mass and engineered as liver organoids to improve their maturity. Encapsulated ProliHHs liver organoids (eLO) were intraperitoneally transplanted to treat liver failure ...

Analgesia for rib fractures: a narrative review
CONCLUSION: Rib fracture(s) continues to be a serious diagnosis, with high rates of mortality, development of chronic pain, and disability. A multidisciplinary approach to management, combined with appropriate analgesia and adherence to care bundles/protocols, has been shown to decrease morbidity and mortality. Most of the risk-stratifying care pathways identified perform poorly in predicting mortality and complications after rib fracture(s).PMID:38459368 | DOI:10.1007/s12630-024-02725-1 (Source: Canadian Journal of Anaesthesia)
