Higher prepubertal IGF-1 concentrations associate to earlier pubertal tempo in both sexes
Introduction: Pubertal onset is triggered by multiple neuroendocrine interactions. The role of prepubertal IGF-1 in this process has not been explored in both sexes. Our objective is to analyze the association of prepubertal IGF-1 concentration with age at thelarche (B2) and menarche (M) in girls and age at gonadarche (G2) in boys. Methods: Longitudinal study (n=1196 boys and girls) within the Growth and Obesity Chilean cohort Study (GOCS). At ages ≈ 6.7 years blood sample for IGF-1. Subjects analyzed into 4 groups according to the onset age of the pubertal event. Results: Higher prepubertal IGF-1 levels were observed ...

Combined Arginine and Insulin Stimulation Elicits a Robust and Consistent Copeptin Response in Short Children
Conclusion: Arginine and insulin appear to have an additive and consistent effect resulting in significant stimulation of copeptin secretion in children. The AITT may be a useful tool to evaluate for normal posterior pituitary function in this age-group, with potential implications for the evaluation of polyuria-polydipsia syndrome.Horm Res Paediatr (Source: Hormone Research in Paediatrics)

Combined arginine and insulin stimulation elicits a robust and consistent copeptin response in short children
The objective of this study was to describe the copeptin response to combined arginine and insulin in children with normal posterior pituitary function. Methods: We conducted a prospective, single-arm assessment of serum copeptin concentrations in children (age 7-16 years, n=38) undergoing growth hormone stimulation testing with an arginine-insulin tolerance test (AITT) for short stature or growth deceleration in a tertiary referral center. After overnight fasting, arginine (500 mg/kg) was administered between 0-30 minutes intravenously (IV) followed by insulin (0.1 units/kg IV) at 60 minutes. Copeptin serum concentration...

Resolved Severe Primary Hypothyroidism in Sensenbrenner Syndrome Post Hepatorenal Transplantation: A Case Report
Conclusion: For the first time in the literature, we report an association of Sensenbrenner syndrome with hypothyroidism that resolved after a combined renal and liver transplant. Such findings expand the clinical spectrum of this syndrome. However, a larger cohort is needed to confirm or exclude such an association. Our case highlights the importance of thyroid function monitoring in any patient with renal and liver failure prior to and after a hepatorenal transplant. KEYWORDS: Sensenbrenner syndrome, cranioectodermal dysplasia, WDR35 gene, hypothyroidism, Cranioectodermal dysplasia 2, liver failure, end-stage renal dise...

Pros and cons of current diagnostic tools for risk-based screening of prediabetes and type 2 diabetes in children and adolescents with overweight or obesity
In conclusion, when choosing the most suitable test in clinical practice, advantages and disadvantages should be considered, as well as the possibility of performing different tests at once. (Source: Hormone Research in Paediatrics)

Treatment of Congenital Hypothyroidism: Impact of Secular Changes in Levothyroxine Initial Dose on Early Growth
Discussion: Current treatment practice with higher LT4 dose normalizes TSH rapidly without significant increase in side effects. However, irrespective of initial LT4 dose, children were shorter than expected at 2 years of age. Effects of different initial LT4 dose on cognitive development urges further investigation. Challenges waiting for an adult with DSD
Disorders/differences of sex development (DSD) comprise a heterogeneous group of inborn conditions where the individual ’s sex chromosomes, gonads and/or anatomical sex are discordant. Since the Chicago consensus conference in 2005, multidisciplinary care has been implemented in specialised pediatric tertiary care centres and clinical practice has substantially changed towards a more holistic approach. Psychologic al support has become a key factor in the management of DSD. After pediatric care, one of the main challenges is the transition of patients to expert care in adulthood. Patients frequently experience difficult...

Effects of a 2-year early childhood vitamin D3 intervention on tooth enamel and oral health at age 6-7 years
Conclusion: Daily supplementation with 10 µg vitamin D3 in the Northern Hemisphere seems adequate in healthy children younger than 2 years in ensuring good oral health at early school age. Trial registration: ClinicalTri als.gov (NCT04302987) (Source: Hormone Research in Paediatrics)

11-Oxygenated Androgen Metabolite Concentrations are Affected by Pubertal Progression and Obesity
Introduction: 11-oxygenated C19 steroids (11-oxyandrogens) have been shown to rise during adrenarche and remain higher throughout adulthood than in early childhood. The patterns of circulating 11-oxyandrogens throughout normal puberty has not yet been described. Methods: We conducted a secondary analysis of healthy youth participants, both males and females, enrolled in six prior endocrine studies (N=249). Participants were classified according to Tanner stage and body mass index (BMI). Concentrations of three adrenal-specific 11-oxygenated androgens, 11β -hydroxyandrostenedione (11OHA4), 11β-hydroxytestosterone (11OHT...
