Functional and phenotypical analysis of IL ‐6‐secreting CD4+ T cells in human adipose tissue
Abstract Emerging evidence indicates that a dynamic interplay between the immune system and adipocytes contributes to the disturbed homeostasis in adipose tissue of obese subjects. Recently, we observed IL‐6‐secretion by CD4+ T cells from the stromal vascular fraction (SVF) of the infrapatellar fat pad (IFP) of knee osteoarthritis patients directly ex vivo. Here we show that human IL‐6+CD4+ T cells from SVF display a more activated phenotype than the IL‐6− T cells, as evidenced by the expression of the activation marker CD69. Analysis of cytokines secretion, as well as expression of chemokine r...

Altered toll ‐like receptor responsiveness underlies a dominant heritable defect in B cell tolerance in autoimmune New Zealand Black mice
Abstract Systemic lupus erythematosus is a debilitating autoimmune disease in which autoantibodies and autoreactive T cells destroy kidneys and other organs. Disease is clinically and genetically heterogeneous, suggesting that underlying mechanisms vary between patients. We previously used an autoantibody transgenic mouse reporter system to examine the effect of different autoimmune backgrounds on B‐cell tolerance, failure of which is a fundamental defect in lupus. We identified a defect consistent with reversible anergy induced by endotoxin stimulation of B cells from Ig transgenic New Zealand Black (NZB) mice. Her...

The gut microbiota: An emerging risk factor for cardiovascular and cerebrovascular disease
Abstract Commensal gut microbiota have recently been implicated in cardiovascular disease (CVD) and cerebrovascular disease. Atherosclerotic plaque formation depends on the colonization status of the host. In addition to host nutrition and the related microbiota‐dependent metabolic changes, activation of innate immune pathways triggers the development of atherosclerosis and supports arterial thrombosis. Gnotobiotic mouse models have uncovered that activation of Toll‐like receptor‐2 by gut microbial ligands supports von Willebrand factor‐integrin mediated platelet deposition to the site of vascular injury. Depending...

Siglec ‐1 inhibits RSV‐induced interferon gamma production by adult T cells in contrast to newborn T cells
In this study, micro‐array data identified the upregulation of sialic acid‐binding immunoglobulin‐type lectin 1 (Siglec‐1) in human RSV‐infected infants. In vitro, RSV increased expression of Siglec‐1 on healthy newborn and adult monocytes. RSV‐induced Siglec‐1 on monocytes inhibited IFN‐γ production by adult CD4+ T cells. In contrast, IFN‐γ production by RSV in newborns was not affected by Siglec‐1. The ligand for Siglec‐1, CD43, is highly expressed on adult CD4+ T cells compared to newborns. Our data show that Siglec‐1 reduces IFN‐γ release by adult T cells pos...

C5aR activation in the absence of C5a: A new disease mechanism of autoimmune hemolytic anemia in mice
Abstract IgG Fc receptors (FcγRs) and the C5a anaphylatoxin receptor (C5aR) were identified as key regulators of type II autoimmune injury in mice. However, and with respect to C5aR, the relative importance of C5a for IgG autoantibody‐induced cellular destruction remained unclear. Using an experimental model of autoimmune hemolytic anemia (AIHA), we here report marked differences in the development of AIHA between mice lacking C5aR and C5‐deficient (Hc0) strain, indicating a limited role of C5 in this type of C5aR‐regulated disease. Ex‐vivo‐analyses of liver homogenates from anemic Hc0 mice demonstrate C5a...

Age ‐associated B cells expanded in autoimmune mice are memory cells sharing H‐CDR3‐selected repertoires
Abstract Age‐associated B cells (ABCs) represent a distinct cell population expressing low levels of CD21 (CD21−/low). The Ig repertoire expressed by ABCs in aged mice is diverse and exhibits signs of somatic hypermutation (SHM). A CD21−/low B‐cell population is expanded in autoimmune diseases, e.g. systemic lupus erythematosus, as well as in lupus‐prone NZB/W mice and in mice lacking a pre‐B cell receptor (SLC−/−). However, the nature of the CD21−/low B cells (hereafter ABCs) in autoimmunity is not well understood. Here we show that in young SLC−/− mice, the...

Inside cover story: Eur. J. Immunol. 1'18
Our inside cover features an immunofluorescence staining of human T cells in contact with lymphoblastoid B cells, which naturally express the IL4I1 enzyme. The B cells were seeded onto polylysine treated glass coverslips before adding allogeneic T cells labeled with a fluorescent dye (red). The conjugates were next permeabilized and labeled with an anti‐IL4I1 antibody revealed by a fluorescent secondary antibody (green). In the image, taken from Aubatin et al. (pp. 106–119), IL4I1 granules secreted by B cells into the immune synaptic cleft, induce global T‐cell inhibition by decreasing the early signaling events ...

In the center panel, the juxtaposition of lung epithelial cells (stained with anti‐E‐cadherin, red) and inflammatory cells (anti‐CD103, green), including migrating DCs and intraepithelial T‐cells, is displayed in addition to nuclear staining (blue). The image is taken from Yoshizawa et al. (pp. 128–150), where the authors show that airway epithelia and adjacent lamina propria locales, not iBALT, represent microenvironments for tissue resident memory T‐cell generation. Front cover story: Eur. J. Immunol. 1'18
Our front cover features an immunofluorescence staining of Bcl‐3 (red) and NF‐κB p50 (green) in psoriasis‐lesion skin. The image relates to the article by Tohyama et al. (pp. 168–179), where the authors show that Bcl‐3 enhances psoriasis‐related gene expression of epidermal keratinocytes in cooperation with p50. IL‐22 and IL‐17A, critical mediators in the pathogenesis of psoriasis, increase nuclear Bcl‐3 and p50 in epidermal keratinocytes. Bcl‐3 might be a future therapeutic target for the treatment of psoriasis. (Source: European Journal of Immunology)
