A miRNome analysis of drug-free manic psychotic bipolar patients versus healthy controls.

A miRNome analysis of drug-free manic psychotic bipolar patients versus healthy controls. Eur Arch Psychiatry Clin Neurosci. 2019 Aug 17;: Authors: Tabano S, Caldiroli A, Terrasi A, Colapietro P, Grassi S, Carnevali GS, Fontana L, Serati M, Vaira V, Altamura AC, Miozzo M, Buoli M Abstract The lifetime presence of psychotic symptoms is associated with more clinical severity, poorer outcome and biological changes in patients affected by bipolar disorder (BD). Epigenetic mechanisms have been evoked to explain the onset of psychotic symptoms in BD as well as the associated biological changes. The main objective of the present study was to evaluate the expression profiles of circulating microRNAs (miRNAs) in drug-free manic psychotic bipolar patients versus healthy controls (HC), to identify possible non-invasive molecular markers of the disorder. 15 drug-free manic psychotic bipolar patients and 9 HC were enrolled and 800 miRNAs expression profile was measured by Nanostring nCounter technology on plasma samples and validated through qPCR. Overall, twelve miRNAs showed a significantly altered expression between the two groups (p

CONCLUSIONS: These findings suggest that smoking initiation and lifetime smoking are likely to be a causal risk factor for developing bipolar disorder. We found some evidence that liability to bipolar disorder increased smoking heaviness. Given that smoking is a modifiable risk factor, these findings further support investment into smoking prevention and treatment in order to reduce mental health problems in future generations. So now, according to the Washington Post, the Trump Administration is “…considering a controversial proposal to study whether mass shootings could be prevented by monitoring mentally ill people for small changes that might foretell violence.”1 As a psychiatrist, I keep wondering how many times the White House will scapegoat those with psychiatric illnesses rather than looking at the scientific data on mass shooters. To understand the problem, consider the case of “Tyler” (a composite of many mass shooter profiles). Tyler is a 19-year-old, single, unemployed high school dropout. He was wo...

Abstract Chromosome conformation capture methods have revealed the dynamics of genome architecture which is spatially organized into topologically associated domains, with gene regulation mediated by enhancer-promoter pairs in chromatin space. New evidence shows that endogenous hormones and several xenobiotics act within circumscribed topological domains of the spatial genome, impacting subsets of the chromatin contacts of enhancer-gene promoter pairs in cis and trans Results from the National Institutes of Health-funded PsychENCODE project and the study of chromatin remodeling complexes have converged to provide ...

Chrisa Hickey’s journey into mental health advocacy started when her son, Tim, was diagnosed with very early onset schizophrenia after being admitted to a psychiatric hospital for the first time at the age of 11.  He had been showing symptoms for years and had received a half dozen different diagnoses. His family was desperately looking for answers. Tim’s illness took a toll on the entire family, which was only exacerbated by the lack of information and resources available to them.  In America, fewer than 100 children per year are diagnosed with very early onset schizophrenia. Chrisa had to find ...

Conclusions: These data suggest that BD drug combination is increasing cholesterol biosynthesis and the newly synthesised cholesterol is being utilised within the cells, possibly for synthesis of new membranes to facilitate neurite outgrowth. This mechanism possibly underpins clinical efficacy in individuals with BD treated with polypharmacy. PMID: 31535581 [PubMed - as supplied by publisher]
