Targeting the BRAF Signaling Pathway in CD133pos Cancer Stem Cells of Anaplastic Thyroid Carcinoma

In this study, we isolated CSCs subpopulation with CD133 surface marker from three ATC cell lines by magnetic cell sorting assay. After confirming the segregation by the flow cytometry method, BRAF and sodium-iodide symporter (NIS) genes were investigated in them before and after incubation with BRAF inhibitor. Also, we evaluated the NIS protein expression and localization. We report FDG PET/CT images of a 63-year-old woman referred for characterization of osteolytic lesions of the cervical spine. This patient with chronic renal failure had a history of follicular thyroid carcinoma with undifferentiated cells, treated by a total thyroidectomy, completed by radioactive iodine in 2017. Because of cancer history, a FDG PET/CT was performed and showed multiple moderately hypermetabolic osteolytic lesions of the spine from C3 to T1, with joint erosion on CT. A laminectomy was performed for cervical spinal cord decompression and revealed a gouty tophus.

CONCLUSIONS: DNA methylation levels of three markers can be useful for differentiating differentiated thyroid cancer from non-malignant follicular thyroid lesions, and may serve as prognostic biomarkers for predicting recurrent or persistent disease following surgery for differentiated thyroid cancer. A 75-year-old lady with previously diagnosed moderate mixed aortic valve disease underwent surveillance transthoracic echocardiography (TTE). This showed raised trans-aortic gradients in the presence of normal aortic valve leaflet motion and turbulent colour flow across a calcified left ventricular outflow tract (LVOT). Therefore, in view of possible subaortic stenosis, she was referred for cardiac computed tomography for further characterisation. Past medical history included total thyroidectomy and 2 rounds of radioiodine ablation for follicular thyroid carcinoma 12 months prior.

We examined the utility of microfluidic digital PCR (dPCR) for detection of BRAF and TERT mutations in thyroid tumors. DNA extracted from 100 thyroid tumors (10 follicular adenomas, 10 follicular cancers, 5 medullary cancers, and 75 papillary thyroid cancer (PTC) were used for detection of BRAF and TERT mutations.
