The extracellular matrix as modifier of neuroinflammation and recovery in ischemic stroke and intracerebral hemorrhage
Neurobiol Dis. 2023 Sep 6:106282. doi: 10.1016/j.nbd.2023.106282. Online ahead of print.ABSTRACTStroke is the second leading cause of death worldwide and has two major subtypes: ischemic stroke and hemorrhagic stroke. Neuroinflammation is a pathological hallmark of ischemic stroke and intracerebral hemorrhage (ICH), contributing to the extent of brain injury but also in its repair. Neuroinflammation is intricately linked to the extracellular matrix (ECM), which is profoundly altered after brain injury and in aging. In the early stages after ischemic stroke and ICH, immune cells are involved in the deposition and remodeling...

Acetylation of p53 in the Cerebral Cortex after Photothrombotic Stroke
Abstractp53 expression and acetylation are crucial for the survival and death of neurons in penumbra. At the same time, the outcome of ischemia for penumbra cells depends largely on the histone acetylation status, but the effect of histone acetyltransferases and deacetylases on non-histone proteins like p53 is largely understudied. With combined in silico and in vitro approach, we have identified enzymes capable of acetylation/deacetylation, distribution, stability, and pro-apoptotic activity of p53 in ischemic penumbra in the course of post-stroke recovery, and also detected involved loci of acetylation in p53. The dynami...

Montelukast Sodium Protects against Focal Cerebral Ischemic Injury by Regulating Inflammatory Reaction via Promoting Microglia Polarization
CONCLUSION: Montelukast sodium can protect against focal cerebral ischemic injury by regulating inflammatory reaction via promoting microglia polarization.PMID:37499731 | DOI:10.1016/j.brainres.2023.148498

Neutralizing RGMa with Elezanumab Promotes Cerebroprotection and Recovery in Rabbit Middle Cerebral Artery Occlusion
AbstractRepulsive guidance molecule A (RGMa) is an inhibitor of neuronal growth and survival which is upregulated in the damaged central nervous system following acute spinal cord injury (SCI), traumatic brain injury, acute ischemic stroke (AIS), and other neuropathological conditions. Neutralization of RGMa is neuroprotective and promotes neuroplasticity in several preclinical models of neurodegeneration and injury including multiple sclerosis, AIS, and SCI. Given the limitations of current treatments for AIS due to narrow time windows to intervention (TTI), and restrictive patient selection criteria, there is significant...

Nonpharmacological Treatments for Hospitalized Patients with Stroke: A Nuanced Approach to Prescribing Early Activity
AbstractStroke remains a leading cause of adult disability. To date, hyperacute revascularization procedures reach 5 –10% of stroke patients even in high resource health systems. There is a limited time window for brain repair after stroke, and therefore, the activities such as prescribed exercise in the earliest period will likely have long-term significant consequences. Clinicians who provide care for hospita lized stroke patients make treatment decisions specific to activity often without guidelines to direct these prescriptions. This requires a balanced understanding of the available evidence for early post-stroke ...

Time course of neuroinflammation after human stroke – a pilot study using co-registered PET and MRI
Microglial activation contributes to both inflammatory damage and repair in experimental ischemic stroke. However, because of the logistical challenges, there have been few clinical imaging studies directly de...

Else_BRB_110660Docosahexaenoic acid promotes M2 microglia phenotype via activating PPAR γ-mediated ERK/AKT pathway against cerebral ischemia-reperfusion injury
Brain Res Bull. 2023 May 4:110660. doi: 10.1016/j.brainresbull.2023.110660. Online ahead of print.ABSTRACTIn ischemia-reperfusion stroke, microglia play a dual role in brain injury as well as brain repair, and promoting their switch from a pro-inflammatory M1 phenotype to an anti-inflammatory M2 phenotype is considered to be a potential therapeutic strategy. Docosahexaenoic acid (DHA) is an essential long-chain omega-3 polyunsaturated fatty acid that exhibits potent anti-inflammatory properties in the acute phase of ischemic stroke, but its effect on microglia polarization is unknown. Thus, the objective of this study was ...

HMGB1, angel or devil, in ischemic stroke
ConclusionsHMGB1 plays a complex role in cerebral infarction, which is related to not only the modification of HMGB1 and bound receptors but also different stages and subtypes of cerebral infarction. future studies on HMGB1 should investigate the spatial and temporal dynamics of HMGB1 after cerebral infarction. Moreover, future studies on HMGB1 should attempt to integrate different stages and infarct subtypes of cerebral infarction.

High prevalence of abdominal aortic aneurysm in older men with cerebrovascular disease: Evaluation of a local screening program
This report evaluates the results of a decade's operation of a local screening program for AAA in this selected neurologic population.MethodsMen aged ≥60 years and admitted to the neurology ward of a community-based hospital in the Netherlands from 2006 to 2017 with a diagnosis of TIA or stroke were selected for screening. The diameter of the abdominal aorta was assessed by abdominal ultrasonography. Patients with detected AAA were referred for evaluation by a vascular surgeon.ResultsAAA was detected in 72 of 1,035 screened patients (6.9%). AAAs with a diameter of 3.0–3.9 cm accounted for 61.1% of the total aneurysms f...

The NG2-glia is a potential target to maintain the integrity of neurovascular unit after acute ischemic stroke
Neurobiol Dis. 2023 Mar 13:106076. doi: 10.1016/j.nbd.2023.106076. Online ahead of print.ABSTRACTThe neurovascular unit (NVU) plays a critical role in health and disease. In the current review, we discuss the critical role of a class of neural/glial antigen 2 (NG2)-expressing glial cells (NG2-glia) in regulating NVU after acute ischemic stroke (AIS). We first introduce the role of NG2-glia in the formation of NVU during development as well as aging-induced damage to NVU and accompanying NG2-glia change. We then discuss the reciprocal interactions between NG2-glia and the other component cells of NVU, emphasizing the factor...
