Clinical introduction A 63-year-old right-handed man presented for evaluation of left leg stiffness and sensory change. He had a medical history of remote prostate cancer (status post resection in 2011). The patient reported that one week prior, he began noticing left leg stiffness and numbness, as well as left hand weakness and discoordination. Neuritin improves the neurological functional recovery after experimental intracerebral hemorrhage in mice
In this study, ICH was induced in C57BL/6 J mice by injection of collagenase VII, while the overexpression of Nrn in the striatum was induced by an adeno-associated virus serotype 9 (AAV9) vector. We found that compared with GFP-ICH mice, Nrn-ICH mice showed improved performance in the corner, cylinder and forelimb tests after ICH, and showed less weight loss and more rapid weight recovery. Overexpression of Nrn reduced brain lesions, edema, neuronal death and white matter and synaptic integrity dysfunction caused by ICH. Genetic Inhibition of Receptor Interacting Protein Kinase-1 Reduces Cell Death and Improves Functional Outcome After Intracerebral Hemorrhage in Mice Basic Sciences
Conclusions—The data show that the kinase domain of RIPK1 is a disease driver of ICH, mediating both acute cell death and functional outcome, and support development of RIPK1 inhibitors as therapeutic agents for human ICH.
