The potential therapeutic effect of phosphodiesterase 5 inhibitors in the acute ischemic stroke (AIS)
In conclusion, PDE5 inhibitors may have potential roles in the management of AIS through modulation of CBF, cAMP/cGMP/NO pathway, neuroinflammation, and inflammatory signaling pathways. Preclinical and clinical studies are recommended in this regard.PMID:37395897 | DOI:10.1007/s11010-023-04793-1

Platelet Function and Sildenafil Use During Left Ventricular Assist Device Support
Clinical outcome studies have shown that sildenafil is associated with reduced mortality and thrombotic events such as ischemic stroke during contemporary left ventricular assist device (LVAD) support. Sildenafil has anti-platelet properties but its relation to platelet function during LVAD support is uncertain.

Phosphodiesterase-5 Inhibitor Attenuates Anxious Phenotypes and Movement Disorder Induced by Mild Ischemic Stroke in Rats
CONCLUSION: PDE5i as effective therapeutic agents for anxiety and movement disorders and provide robust preclinical evidence to support the development and use of PDE5i for the treatment of mild ischemic stroke residual disorders.PMID:35430790 | DOI:10.3340/jkns.2021.0101

Treprostinil palmitil inhibits the hemodynamic and histopathological changes in the pulmonary vasculature and heart in an animal model of pulmonary arterial hypertension
In this study, TP was delivered by inhalation (treprostinil palmitil inhalation suspension, TPIS) in a rat sugen 5416 (Su)/hypoxia (Hx) model of pulmonary arterial hypertension (PAH) to evaluate its effects on hemodynamics, pulmonary vascular remodeling, and cardiac performance and histopathology. Male Sprague-Dawley rats received Su (20 mg/kg, s.c), three weeks of Hx (10% O2) and 5 or 10 weeks of normoxia (Nx). TPIS was given during the 5-10 week NX period after the Su/Hx challenge. Su/Hx increased the mean pulmonary arterial blood pressure (mPAP) and right heart size (Fulton index), reduced cardiac output (CO), stroke vo...

Right ventricular and cyclic guanosine monophosphate signalling abnormalities in stages B and C of heart failure with preserved ejection fraction
ConclusionExercise CMR identifies impaired RV-arterial coupling at an early stage of HFpEF. Circulating cGMP levels phenocopy the haemodynamic spectrum in HFpEF but fail to increase after phosphodiesterase type 5 inhibition, endorsing the need for alternative interventions to increase cGMP signalling in HFpEF.

Sildenafil Administration Improves Right Ventricular Function on 4D Flow MRI in Young Adults Born Premature
CONCLUSIONS: Despite reduced right atrial workload, metoprolol significantly depressed overall cardiac systolic function. Sildenafil, however, increased CI and improved RV function, as quantified by the direct flow fraction. The preterm heart appears dependent on HR, but sensitive to RV afterload manipulations.PMID:33861148 | DOI:10.1152/ajpheart.00824.2020
