K < sub > V < /sub > 4 channels in isolectin B4 muscle dorsal root ganglion neurons of rats with experimental peripheral artery disease: Effects of bradykinin B1 and B2 receptors
In conclusion, our data suggest that limb ischemia and/or ischemia induced-BK inhibit activity of KV4 channels in a subpopulation of the thin fiber muscle afferent neurons depending on GDNF, which is likely a part of signaling pathways involved in the exaggerated blood pressure response during activation of muscle afferent nerves in PAD.PMID:36094447 | DOI:10.1152/ajpregu.00117.2022

The hypoxia-inducible factor- α prolyl hydroxylase inhibitor FG4592 ameliorates renal fibrosis by inducing the H3K9 demethylase JMJD1A
Am J Physiol Renal Physiol. 2022 Sep 8. doi: 10.1152/ajprenal.00083.2022. Online ahead of print.ABSTRACTHypoxia-inducible factors 1α and 2α (HIF-1α/2α) transcription factors are the major regulators of the cellular response to hypoxia and also play a key role in renal fibrosis associated with acute and chronic kidney disease. Jumonji domain-containing 1a (JMJD1A), a histone H3 lysine 9 (H3K9) demethylase, is reported to be an important target gene of HIF-α. However, whether JMJD1A and H3K9 methylation status play a role in renal fibrosis is unclear. Here, we investigated the involvement of HIF-α, JMJD1A, and monometh...

Electroacupuncture pre-treatment alleviates sepsis-induced cardiac inflammation and dysfunction by inhibiting the calpain-2/STAT3 pathway
In this study, we aimed to elucidate the mechanism by which electroacupuncture reduces cardiac inflammation and apoptosis and improves cardiac function during sepsis. Electroacupuncture pretreatment for 7 days was applied in septic cardiomyopathy model induced by lipopolysaccharide (LPS). lipopolysaccharide-induced sepsis was associated with a dramatically systemic inflammation and cardiac dysfunction, which was alleviated by electroacupuncture pre-treatment. Lipopolysaccharide resulted in increases of pro-inflammatory factors (TNF-α,IL1βand IL-6) and apoptosis (TUNEL staining and BAX/Bcl2) via activation of calpain-2/S...

β-PIX cooperates with GIT1 to regulate eNOS in sinusoidal endothelial cells
CONCLUSIONS: The data indicate that b-PIX regulates eNOS phosphorylation and function in normal SECs, and highlight the importance of the GIT1/b-PIX/eNOS trimolecular complex in normal liver SEC function.PMID:36044673 | DOI:10.1152/ajpgi.00034.2022

Lactate-induced lactylation in skeletal muscle is associated with insulin resistance in humans
This study identified the existence of a lactate-derived post-translational modification in human skeletal muscle and suggests skeletal muscle lactylation could provide additional insight into the regulation of skeletal muscle metabolism, including insulin resistance.

Protections of transcription factor BACH2 and natural product myricetin against pathological cardiac hypertrophy and dysfunction
This study aims to explore the role of BACH2, a member of the basic region leucine zipper transcription factor family, in cardiac hypertrophy and failure. Transverse aortic constriction surgery was operated to induce cardiac hypertrophy and failure in mice. BACH2 was overexpressed in mice through tail vein injection of AAV9-Bach2. Mice with systemic or cardiac-specific knockdown of Bach2 were adopted. Neonatal rat ventricular myocytes (NRVMs) were isolated and infected with lentivirus to overexpress Bach2 or transfected with siRNA to knock down Bach2. Our data showed that overexpression of BACH2 ameliorated TAC-induced car...

NF- κB-dependent repression of Sox18 transcription factor requires the epigenetic regulators histone deacetylases 1 and 2 in acute lung injury
In acute lung injury (ALI), the NF-κB-mediated downregulation of Sox18 gene expression leads to the disruption of the pulmonary endothelial barrier. Previous studies have suggested that the action of NF-κB as a transcriptional repressor also requires the action of class I histone deacetylases (HDACs). Thus, the purpose of this study was to investigate and further delineate the mechanism of Sox18 repression during lipopolysaccharide (LPS) induced ALI. Using selective inhibitors and specific siRNA-driven depletion of HDACs 1-3 in human lung microvascular endothelial cells (HLMVEC) we were able to demonstrate a critical rol...

Loss of sphingosine kinase 2 protects against cisplatin induced-kidney injury
Am J Physiol Renal Physiol. 2022 Jul 14. doi: 10.1152/ajprenal.00229.2021. Online ahead of print.ABSTRACTCisplatin is an established chemotherapeutic drug for treatment of solid-organ cancers, and is the primary drug utilized in the treatment of head and neck cancer; however, cisplatin-induced nephrotoxicity largely limits its clinical use. Inhibition of sphingosine kinase 2 (SphK2) has been demonstrated to alleviate various kidney diseases. Therefore, we hypothesized that inhibition of SphK2 could also protect against cisplatin-induced nephrotoxicity. Results from the present study showed that the SphK2 inhibitor, ABC2946...
