TAT decorated siRNA polyplexes for inhalation delivery in anti-asthma therapy
Eur J Pharm Sci. 2023 Sep 16;190:106580. doi: 10.1016/j.ejps.2023.106580. Online ahead of print.ABSTRACTIn this work, a novel protonable copolymer was designed to deliver siRNA through the inhalation route, as an innovative formulation for the management of asthma. This polycation was synthesized by derivatization of α,β-poly(N-2-hydroxyethyl)D,L-aspartamide (PHEA) first with 1,2-Bis(3-aminopropylamino)ethane (bAPAE) and then with a proper amount of maleimide terminated poly(ethylene glycol) (PEG-MLB), with the aim to increase the superficial hydrophilicity of the system, allowing the diffusion trough the mucus layer. On...

Luteolin alleviated neutrophilic asthma by inhibiting IL-36 γ secretion-mediated MAPK pathways
CONCLUSIONS: Luteolin alleviated neutrophilic asthma by inhibiting IL-36γ secretion-mediated MAPK pathways. These findings provided a theoretical basis for the application of luteolin in the treatment of neutrophilic asthma.PMID:36604842 | DOI:10.1080/13880209.2022.2160770

Current Drug Targeting using siRNAbased Nano Therapeutics for Pulmonary Diseases
Curr Pharm Des. 2022 Jul 27. doi: 10.2174/1381612828666220727144223. Online ahead of print.ABSTRACTThe importance of siRNA in nano drug delivery systems to target important pulmonary disorders such as chronic obstructive pulmonary disease (COPD), asthma, lung cancer, and others is reviewed in this perspective. The great majority of lung illnesses are caused by protein misfolding. As a result, siRNA-based therapies are increasingly being used to target the gene. Given the difficulties of delivering bare siRNA, siRNA protection may ensure its efficacy in gene therapy. These issues could be solved with a nano-based siRNA deli...

Pref-1 induced lung fibroblast differentiation by hypoxia through integrin α5β1/ERK/AP-1 cascade
Eur J Pharmacol. 2021 Jul 28:174385. doi: 10.1016/j.ejphar.2021.174385. Online ahead of print.ABSTRACTChronic obstructive asthma is characterized by airway fibrosis. Hypoxia and connective tissue growth factor (CTGF) play important roles in airway fibrosis. Preadipocyte factor-1 (Pref-1) participates in adipocyte differentiation and liver fibrosis. Herein, we investigated the role of Pref-1 in airway fibrosis in chronic obstructive asthma. We found that Pref-1 was overexpressed in lung tissues from chronic obstructive asthma patients compared to normal subjects. Extracellular matrix proteins were inhibited by Pref-1 small ...

Abnormal ADAM17 expression causes airway fibrosis in chronic obstructive asthma
Biomed Pharmacother. 2021 May 26;140:111701. doi: 10.1016/j.biopha.2021.111701. Online ahead of print.ABSTRACTPatients with chronic obstructive asthma (COA) develop airflow obstruction caused by subepithelial fibrosis. Although a disintegrin and metalloproteinase 17 (ADAM17) has been implicated in lung inflammation and tissue fibrosis, its role in airway fibrosis in COA has not been explored. Here, we found marked overexpression of ADAM17, phosphorylated ADAM17, and connective tissue growth factor (CTGF) in human airway fibroblasts from COA patients, compared with those of normal subjects. Similarly, levels of ADAM17, CTGF...

Effect of TLR7 gene expression mediating NF- κB signaling pathway on the pathogenesis of bronchial asthma in mice and the intervention role of IFN-γ.
CONCLUSIONS: Upregulation of TLR7 can inhibit the activation of NF-κB signaling pathway, reduces airway inflammation, inhibits ASMCS proliferation and thus promotes cell apoptosis in asthmatic mice. Besides, IFN-γ can exert a protective role in suppressing the progression of inflammation in asthma. PMID: 33577041 [PubMed - as supplied by publisher]

S1PR2 Inhibition Attenuates Allergic Asthma Possibly by Regulating Autophagy
This study is to investigate the role of Sphingosine-1-phosphate (S1P) in the asthma progression, and the involvement of autophagy. Airway remodeling mice were subjected to the HE, PAS, and Masson staining. Protein expression levels in the tissues, samples and model cells were detected with ELISA, Western blot analysis, and immunohistochemical/immunofluorescent analysis. The S1P2 receptor antagonist JTE-013 decreased the inflammatory cell infiltration and goblet cell production in asthmatic mice tissues. The IL-1, IL-4, IL-5 and serum IgE contents were decreased in bronchoalveolar lavage fluid, while the Beclin1 expression...

siRNA delivery to macrophages using aspherical, nanostructured microparticles as delivery system for pulmonary administration.
Abstract The delivery of oligonucleotides such as siRNA to the lung is a major challenge, as this group of drugs has difficulties to overcome biological barriers due to its polyanionic character and the associated hydrophilic properties, resulting in inefficient delivery. Especially in diseases such as asthma, chronic obstructive pulmonary disease and cystic fibrosis, where increased proinflammation is present, a targeted RNA therapy is desirable due to the high potency of these oligonucleotides. To address these problems and to ensure efficient uptake of siRNA in macrophages, a microparticulate, cylindrical deliv...
