Role of Fibulin 3 in Aging ‐Related Joint Changes and Osteoarthritis Pathogenesis in Human and Mouse Knee Cartilage
ConclusionFibulin 3 is specifically expressed in the SZ of articular cartilage and its expression is reduced in aging and OA. Fibulin 3 regulates differentiation of adult progenitor cells, and its aging‐related decline is an early event in the pathogenesis of OA. Preventing aging‐associated loss of fibulin 3 or restoring it to normal levels in SZ chondrocytes has the potential to delay or prevent the onset of OA.

ConclusionThese findings implicate miR‐9–mediated suppression of MCPIP‐1 in the pathogenesis of OA via up‐regulation of IL‐6 expression in IL‐1β–stimulated human OA chondrocytes.

Monocarboxylate transporter 4, associated with the acidification of synovial fluid, is a novel therapeutic target for inflammatory arthritis
Conclusion. RA activity correlated with decreased synovial fluid pH. This may be due to increased MCT4 expression in RASFs. Silencing MCT4 induced apoptosis in RASFs and reduced the severity of CIA, suggesting that MCT4 is a potential therapeutic target for inflammatory arthritis. This article is protected by copyright. Interleukin‐1 Acts via the JNK‐2 Signaling Pathway to Induce Aggrecan Degradation by Human Chondrocytes
ConclusionThe signaling axis TRAF‐6/TAK‐1/MKK‐4/JNK‐2 mediates IL‐1–induced aggrecanolysis. The level of aggrecanase is controlled by its endocytosis, which may be reduced upon IL‐1 stimulation because of LRP‐1 shedding.

ConclusionAnnexin A2 is a key molecule in the DDR‐2/annexin A2/MMP‐13 loop, the activation of which contributes to joint destruction in RA, mainly through promoting invasion of FLS. Annexin A2 might therefore become a novel clinical target for RA treatment.

Stress inducible gene nuclear protein‐1 (NUPR1/p8) regulates MMP‐13 expression in human articular chondrocytes
Conclusions: Nupr1 is expressed in cartilage, and its levels are increased in OA. Nupr1 expression is required for IL‐1β‐mediated expression of MMP‐13. Thus, our study suggests a novel pathway for regulation of IL‐1β‐mediated production of MMPs in chondrocytes. © 2014 American College of Rheumatology.

Long noncoding RNA related to cartilage injury promotes chondrocyte extracellular matrix degradation in osteoarthritis

Class I Histone Deacetylase Inhibition Modulates Metalloproteinase Expression and Blocks Cytokine‐Induced Cartilage Degradation
ConclusionInhibition of class I HDACs (HDAC‐1, HDAC‐2, HDAC‐3) by MS‐275 or by specific depletion of HDACs is capable of repressing cytokine‐induced metalloproteinase expression in cartilage cells and BNC explants, resulting in inhibition of cartilage resorption. These observations indicate that specific inhibition of class I HDACs is a possible therapeutic strategy in the arthritides.
