Cyclophilin D Contributes to Airway Epithelial Mitochondrial Damage in Chronic Obstructive Pulmonary Disease
ConclusionThese data suggest that CypD signaling pathway is involved in the pathogenesis of COPD and provide a potential therapeutic target for COPD.

Cigarette smoking is a secondary cause of folliculin loss
Conclusions Our study demonstrates that CS exposure is a secondary cause of FLCN deficiency due to the enhanced proteolysis, which promoted airway epithelial cell death.

MiR-125b-5p up-regulation in asthma and COPD overlap patients mediates oxidative stress and apoptosis via targeting IL6R/NLRC5/TRIAP1
Conclusions: MiR-125b-5p is up-regulated in ACO patients, and may cause oxidative stress and cell apoptosis through targeting IL6R, NLRC5, TRIAP1 genes.

Role of urokinase-type plasminogen activator receptor (uPAR) in senescent lung cells
Conclusion: uPAR is highly expressed in lung fibroblasts but not increased in senescence. In SAEC, uPAR may be elevated in senescence but does not correlate with the senescence markers examined. This suggests that uPAR is unlikely to be a useful target for senescent lung cell removal.

The role of retinoid-related orphan receptor- α in cigarette smoke-induced autophagic response
CONCLUSIONS: Our findings suggest that RORα promotes autophagy and contributes to COPD pathogenesis via regulation of the RORα-p53-DRAM pathway.PMID:35509068 | DOI:10.1186/s12931-022-02034-5

LL-37 and HMGB1 induce alveolar damage and reduce lung tissue regeneration via RAGE
Am J Physiol Lung Cell Mol Physiol. 2021 Aug 18. doi: 10.1152/ajplung.00138.2021. Online ahead of print.ABSTRACTThe receptor for advanced glycation end-products (RAGE) has been implicated in the pathophysiology of chronic obstructive pulmonary disease (COPD). However, it is still unknown whether RAGE directly contributes to alveolar epithelial damage and abnormal repair responses. We hypothesize that RAGE activation not only induces lung tissue damage but also hampers alveolar epithelial repair responses. The effects of the RAGE ligands LL-37 and HMGB1 were examined on airway inflammation and alveolar tissue damage in wild...

Epithelium-derived IL17A Promotes Cigarette Smoke-induced Inflammation and Mucus Hyperproduction
In this study, we aim to determine whether epithelial-derived IL17A regulates inflammation and mucus hyperproduction in COPD using a cultured human bronchial epithelial (HBE) cell line in vitro and airway epithelium IL17A-specific knockout mouse in vivo. Increased IL17A expression was observed in mouse airway epithelium upon cigarette smoke (CS) exposure or in a COPD mouse model that was induced by CS and elastin. CS extract (CSE) also triggered IL17A expression in HBE cells. Blocking IL17A or IL17RA effectively attenuated CSE-induced MUC5AC and the inflammatory cytokines IL6, tumor necrosis factor (TNF)-α, and IL1β in H...

Cigarette Smoke Activates NOTCH3 to Promote Goblet Cell Differentiation in Human Airway Epithelial Cells.
The objective of this study was to determine whether NOTCH signaling regulates goblet cell differentiation in response to cigarette smoke. Primary human bronchial epithelial cells (HBECs) from nonsmokers and COPD smokers were differentiated in vitro on air-liquid interface and exposed to cigarette smoke extract (CSE) for 7 days. NOTCH signaling activity was modulated using (1) the NOTCH/γ-secretase inhibitor Dibenzazepine (DBZ), (2) lentiviral over-expression of the NOTCH3-intracellular domain (NICD3) or (3) NOTCH3-specific siRNA. Cell differentiation and response to CSE were evaluated by qPCR, Western blotting, immunosta...
